摘要
普通光学显微镜受光源衍射现象导致分辨率受限,难以对生物样本进行高分辨率成像。近些年来,发展了一种利用光学显微镜结合膨胀水凝胶的超分辨率光学显微技术——膨胀显微技术(expansion microscopy,ExM)。该技术采用可膨胀的水凝胶均匀地物理拓展生物样本来实现超分辨率。ExM允许在常规光学显微镜下对生物样品进行纳米级成像,包括成功可视化蛋白质、核酸等生物分子,并具有用于临床样本进行病理检查的潜力。在本综述中介绍了ExM的各种方案及其在生物医学领域的应用。
Ordinary optical microscope is limited by the imaging magnification,and it is difficult to image biological samples with high resolution.In recent years,a super-resolution optical microscopy technique using optical microscopy combined with an expanded hydrogels has been developed-(Expansion Microscopy,ExM).The technique uses expandable hydrogels to physically expand biological samples in a isotropic manner to achieve super-resolution.ExM allows nanoscale imaging of biological samples under conventional optical microscopy,including the successful visualization of biomolecules such as protein,nucleic acid,and has the potential for pathological examination of clinical samples.This paper reviews various approaches of ExM and its applications in biomedical field.
