黄肝煎液中主要成分的鉴别与含量测定 被引量:1

摘要 目的建立黄肝煎液的质量控制方法。方法采用薄层色谱(TLC)法对黄肝煎液中的当归及大黄进行鉴别;采用双波长高效液相法同时测定黄肝煎液中绿原酸、黄芩苷、大黄素及大黄酚的含量。YMC-Pack Pro C18色谱柱(250 mm×4.6 mm,5μm);大黄素及大黄酚的检测波长为254 nm,黄芩苷、绿原酸的检测波长为327 nm;流动相为乙腈(A)-0.1%磷酸水(B)梯度洗脱;流速:1.0 mL·min^-1;进样量:20μL;柱温:35℃。结果TLC中的斑点清晰,易于识别,阴性对照无干扰;高效液相含量测定,4种成分在各自考察的浓度范围内,线性关系良好(r>0.9993)。结论该方法专属性强,操作简单,结果准确,可用于该制剂的质量控制。 Objective To establish a method of quality control for Huanggan decoction.Methods Angelica sinensis and Rhei Radix et Rhizoma in Huanggan decoction were qualitatively identified by Thin-layer Chromatography(TLC)method.A double wavelength High Performance Liquid Chromatography(HPLC)method was used for the simultaneous determination of chlorogenic acid,baicalin,emodin and chrysophanol in Huanggan decoction.The analysis was performed on a YMC-Pack Pro C18 column(250 mm×4.6 mm,5μm)with gradient elution using acetonitrile-0.1%phosphoric acid as mobile phase in a flow rate of 1.0 mL·min^-1.Emodin and chrysophanol were detected at 254 nm,chlorogenic acid and baicalin were detected at 327 nm.The temperature of column was set at 35℃,and volume of injection was 20μL.Results The spots in TLC were clear and easily distinguished without interference in negative control.The results of content determination showed that 4 components with good linear relationship in range of respective concentrations(r>0.9993).Conclusion The method is sensitive,simple,and accurate.It can be used for the quality control of the Huanggan decoction.
