摘要
目的采用包载阿霉素的碳酸钙纳米颗粒研究对骨肉瘤的抑制作用,通过碳酸钙的酸性释放药物达到阿霉素的肿瘤定点释放,提高化疗效果。方法通过纳米材料的研制能够很好地解决化疗药物在骨肉瘤组织不敏感、全身循环的难点,利用5~6周雄性质量约25 g的BALB/c裸鼠,分为PBS组、DOX组、DOX@CaCO3组3组,构建骨肉瘤模型,通过给药后观察小动物疗效。结果通过治疗后,DOX@CaCO3组肿瘤生长速率明显低于PBS组及DOX组,肿瘤的大小也明显变小。结论纳米载体系统可实现药物在肿瘤组织定点释放或激活,提高局部药物浓度,减少正常组织的药物蓄积,降低不良反应。
Objective To study the inhibitory effect of calcium carbonate nanoparticles coated with adriamycin on osteosarcoma.Methods The nanomaterials can effectively solve the difficulties of insensitive chemotherapy drugs in osteosarcoma tissues and systemic circulation.BALB/c male nude mice with a male weighing about 25 g and aging 5-6 weeks were divided into PBS,DOX and DOX@CaCO3 groups.The osteosarcoma model was constructed to observe the efficacy.Results After treatment,the tumor growth rate of DOX@CACO3 group was significantly lower than those of PBS group and DOX group,and the tumor size was also significantly reduced.Conclusion Nanocarrier system coated with drug can targeting drug release in tumor tissues,increase local drug concentration,reduce drug accumulation in normal tissues and reduce adverse reactions.
