摘要
以东亚飞蝗卵为材料,直接取其胚胎细胞涂片、染色,进行染色体观察,并对东亚飞蝗染色体核型进行了初步分析。结果表明:孵化3~4 d的虫卵是较佳的取样时相点,采用该方法制片简便、省时,得到的标本染色体分散度良好、形态清晰,分裂相多,有丝分裂时期完整。该制片方法的建立不仅为昆虫核型研究和染色体行为分析提供了便捷的手段,同时也为染色体相关的实践教学提供一种简便易学的参考方案。
In the study,the eggs of Locusta migratoria were used as materials,and the embryonic cells of the eggs were directly collected for smear and staining for chromosome observation. A preliminary analysis of the karyotypes of Locusta migratoria was carried out. The results showed that the 3-4 days of hatching eggs were the preferred sampling points,the method was simple and timesaving,and the obtained samples had good chromosome dispersion,clear shape,many mitotic figures,and the mitosis process was complete. The establishment of this method not only serves as a convenient means for insect karyotype research and chromosome behavior analysis but also provides a simple and easy method to learn a reference program for chromosome-related practice teaching.
