摘要
对大鲵肉进行酶解制备具有抗氧化和免疫性的活性肽,以提高大鲵肉的保健功能。分别利用碱性蛋白酶、风味蛋白酶和胰蛋白酶对其水解制备酶解肽,测定不同酶解肽的免疫活性和抗氧化性。结果表明,3种酶在适宜条件下酶解3 h后,酶解肽的水解度分别为28.55%、10.36%和15.58%;分子量在2 kDa以下分别为98.85%、93.36%和98.22%。3种酶解肽DPPH自由基清除能力,IC 50值分别为5.92、3.54、5.43 mg/mL;超氧阴离子自由基清除能力,IC 50值分别为0.38、4.87、1.96 mg/mL;亚铁离子螯和能力,IC 50值分别为1.06、1.14、1.39 mg/mL;3种酶解肽浓度400μg/mL对免疫细胞增殖率分别为17.96%±0.49%、35.97%±1.56%和16.58%±0.78%。风味蛋白酶酶解肽抗氧化和免疫活性显著高于其它酶解。
Enzymatic hydrolysis of Chinese giant salamander meat has been conducted to prepare peptides with high antioxidant and immunological activity.The enzymatic peptides have been prepared by hydrolysis with alkaline protease,flavor protease and trypsin,respectively,and the immunological activity and antioxidant activity of different enzymatic peptides have been determined.The results show that the hydrolysis degree of the enzymatic peptides is 28.55%,10.36%and 15.58%after the three enzymes are hydrolyzed under suitable conditions,and the molecular weights are 98.85%,93.36%and 98.22%below 2 kDa,respectively.The DPPH free radical scavenging ability of the three enzymatic peptides is 5.92,3.54,and 5.43 mg/mL,respectively;with the superoxide anion radical scavenging ability,IC 50 values are 0.38,4.87,1.96 mg/mL,respectively;with ferrous ion chelation,the IC 50 values are 1.06,1.14,1.39 mg/mL,and the proliferation rates of the three enzymatic peptides at 400μg/mL are 17.96%±0.49%,35.97%±1.56%,and 16.58%±0.78%respectively.The anti-oxidation and immunological activities of the flavor protease hydrolyzing peptide are significantly higher than other enzymatic hydrolysis.
