摘要
目的研究连翘挥发油自微乳处方组成和比例,并考察其解热药效。方法通过溶解性实验、因素考察和伪三相图绘制,以自乳化时间、乳化效果、乳滴粒径、多分散系数、稳定性为指标确定最佳处方;建立酵母致热大鼠模型,考察其和挥发油在体内解热药效。结果连翘挥发油自微乳的处方组成为连翘挥发油-油酸乙酯-吐温20-乙醇(25∶10∶81∶9),制得外观澄清透明的淡黄色油状液体,粒径为(37.41±0.014)nm,多分散系数0.211±0.018,电位(-7.05±0.53)mV,乳化时间32 s。解热药效结果显示,连翘挥发油自微乳能够有效抑制发热模型大鼠体温上升,降低模型大鼠体温。结论连翘挥发油自微乳制备简单,增加了药物在水中溶解度,有利于胃肠道吸收,提高了挥发油解热药效,为连翘挥发油新剂型的开发与利用奠定基础。
Objective To study the composition and proportion of Forsythia suspense volatile oil for oral self-microemulsifying drugdelivery system and investigate its antipyretic effect. Methods The prescription and preparation technology were selected and optimized through the solubility experiment,factortestand pseudo-ternaryphase diagram method, using the self-emulsifying time, appearance, particle diameter, PDI and stability as selection indexes. We investigated its antipyretic efficacy in vivo by establishing a dry yeast-induced rat model.Result The prescription composition of Forsythia suspense volatile oil self microemulsion wasForsythia volatile oil-ethyloleate-Tween20-ethanol(25∶10∶40∶9). The acquired Forsythia suspense volatile oil self-microemulsion exhibited uniform and transparent, with the particle diameter(37.41±0.014)nm, polydispersity index 0.211±0.018, Zeta potential(-7.05±0.53) mV, self-emulsifying time 38 s. The result showed Forsythia suspense volatile oil self-microemulsion can effectively inhibit the temperature increase of rats with fever and decrease the temperature instead. Conclusion The preparation of Forsythia suspense volatile oil self-microemulsion is simple, increasing the solubility in water, making it better absorption in the stomach and intestine and enhancing the efficacy of antipyretic effect. It provides a basis for further development and utilization of Forsythia suspense volatile oil.
