摘要
目的探究补肝敛肺汤对酒精性损伤大鼠氧化应激及炎性反应的影响。方法2018年2月—2019年4月将60只SD大鼠随机分为3组,分别为对照组、模型组和补肝敛肺汤组,每组20只SD大鼠,补肝敛肺汤组分别通过灌胃的方式给药(模型组灌胃予以相同剂量的生理盐水),持续时间30 d,而后均灌胃予以白酒5 g/kg,对照组大鼠灌胃予以等剂量生理盐水,于干预30 d后检测大鼠血清样本中丙氨酸氨基转移酶(alanine transaminase,ALT)、天冬氨基酸氨基转移酶(aspartate aminotransferase,AST)、血清总胆固醇(total cholesterol,TC)、总胆红素(total bilirubin,TBIL)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子(tumor necrosis factor,TNF-α)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)水平,并观察不同组别大鼠肝脏病理切片进行比较;结果(1)病理切片观察发现,对照组大鼠肝脏组织肝小叶结构正常,无变性坏死,模型组大鼠肝细胞瘀血肿胀,出现炎性细胞浸润,补肝敛肺汤组大鼠肝脏细胞病变情况较模型组有改善;(2)对比显示,模型组大鼠ALT、AST、TC、ALB均较对照组出现明显升高(P<0.05),补肝敛肺汤组大鼠上述指标水平较模型组有下降(P<0.05),但高于对照组(P<0.05);(3)模型组大鼠IL-6及TNF-α水平明显高于对照组(P<0.05),补肝敛肺汤组大鼠上述指标水平较模型组有下降(P<0.05),但高于对照组(P<0.05);(4)模型组大鼠SOD(超氧化物歧化酶)水平明显低于对照组(244.26±19.68)vs(401.23±20.15),MDA(丙二醛)水平明显高于对照组(29.65±5.23)vs(11.81±3.02)(P<0.05),补肝敛肺汤组大鼠SOD水平高于模型组(320.15±16.55)vs(244.26±19.68),MDA水平低于模型组(20.36±3.65)vs(29.65±5.23)(P<0.05)。结论补肝敛肺汤对酒精性肝损伤大鼠具有较好的治疗效果,能够显著降低大鼠氧化应激反应及炎性反应。
Objective To investigate the effects of buganliongfei decoction on oxidative stress and inflammatory response in rats with alcohol injury.Methods 60 SD rats from February 2018 to April 2019 were randomly divided into three groups,control group,model group respectively and tonifying liver folding of the lung decoction group,each group with 20 SD rats,tonifying liver folding of the lung decoction group respectively by means of filling the stomach medication(model group to fill the stomach to the same dose of saline),duration of 30 d,then all lavage liquor 5 g/kg,control group rats lavage isodose physiological saline,alanine aminotransferase(ALT),aspartame amino acid aminotransferase(AST),serum total cholesterol(TC),total bilirubin(TBIL),interleukin-6(IL-6),tumor necrosis factor(TNF-α),malondialdehyde and superoxide dismutase(SOD)were detected in rat serum samples after 30 days of intervention.Pathological sections of liver of different groups of rats were observed and compared.Results(1)pathological observation showed that the liver lobule structure of the control group was normal without degeneration and necrosis,while the model group showed liver cell stasis and swelling with inflammatory cell infiltration.(2)comparison showed that ALT,AST,TC and ALB in the model group were significantly higher than those in the control group(P<0.05),while the levels of the above indicators in the rats in the buliangliongfei decoction group were significantly lower than those in the model group(P<0.05),but higher than those in the control group(P<0.05).(3)the level of IL-6 and TNF-in rats in the model group was significantly higher than that in the control group(P<0.05),and the level of the above indicators in rats in the buliangliongfei decoction group decreased compared with that in the model group(P<0.05),but was higher than that in the control group(P<0.05).(4)SOD level in the model group was significantly lower than that in the control group(244.26±19.68 vs 401.23±20.15),and MDA level was significantly higher than that in the control group(29.65±5.23 vs 11.81±3.02)(P<0.05).Conclusion Buliangliongfei decoction has a good therapeutic effect on rats with alcoholic liver injury,and can significantly reduce oxidative stress and inflammatory reaction in rats.
