山茶油中原花青素的提取工艺优化及含量测定 被引量:9

摘要 优化山茶油中原花青素的提取工艺,并建立其含量测定方法。以原花青素标准品为对照品,采用紫外分光光度法测定山茶油中原花青素的含量。通过单因素试验和正交试验相结合的方法考察甲醇浓度、提取温度、提取时间和提取次数对山茶油中原花青素提取的影响。山茶油中原花青素最佳提取工艺为甲醇体积分数50%、提取温度80℃、提取时间40 min、提取4次;在此条件下所提取得到山茶油中平均原花青素含量为265.90 mg/kg。山茶油中含有较高含量的原花青素,所得的优选工艺稳定可行,原花青素含量测定方法可靠。 To optimize the extraction technology of procyanidins from camellia oil and establish its content determination method.The content of procyanidins in camellia oil was determined by UV spectrophotometry with the standard procyanidins as the reference.The effects of methanol concentration,extraction temperature,extraction time and extraction times on the extraction of procyanidins from camellia oil were investigated by single factor test and orthogonal test.The optimal extraction technology conditions of procyanidins in camellia oil were as follows:methanol volume fraction 50%,water-bath temperature 80℃,extraction time 40 min and extraction for 4 times.Under these conditions,the average content of procyanidins in camellia oil was 265.90 mg/kg.Camellia oil contained high content of procyanidins,the optimized technology was stable and feasible,and the method for determination of procyanidins was reliable.
