摘要
Wnt/β-catenin信号通路在小鼠舌发育的过程中起着重要的作用,但是在人舌早期发育过程中的作用研究未见报道.该研究以人舌胚为材料,通过RNA原位杂交技术和Real-time PCR技术检测Wnt/β-catenin信号通路的关键因子(配体WNT3,受体FZD4,转录因子TCF4,抑制因子DKK1)在人舌胚早期发育过程中的表达模式.结果显示,WNT3、FZD4、TCF4和DKK1基因均在9周和12周人舌胚的上皮和舌肌中表达,而在舌间充质未见表达.利用免疫荧光技术验证结果显示,Wnt/β-catenin信号通路的关键转导因子(CTNNB1蛋白)在人舌胚的上皮表达.研究表明,Wnt/β-catenin信号通路的关键因子WNT3、FZD4、TCF4、DKK1和关键转导因子CTNNB1通过在舌上皮表达参与调控人舌胚发育过程.
Wnt/β-catenin signaling components play an important role in the development of mouse tongue, but little is known about the situation of these molecules in human embryonic tongue. In this study, expression patterns of Wnt/β-catenin signaling pathway critical factors(WNT ligand WNT3,receptor FZD4, transducer TCF4 and antagonist DKK1) were detected in early development human tongue by in situ hybridization. The results showed that WNT3, FZD4,TCF4 and DKK1 were examined in the epitheliums and muscles of tongue,in which WNT3 and DKK1 showed slightly stronger localization.And then,CTNNB1 protein was also detected in the epitheliums of the tongue by immunofluorescence. The results revealed the status of active WNT3,FZD4,TCF4,DKK1 and CTNNB1 signaling in the developing human tongue.
