摘要
【目的】蔗糖转化酶是植物中糖信号调控途径中的关键因子之一,它们在调控植物糖代谢、生长发育、抗病和抗逆等过程中起着至关重要的作用。蔗糖转化酶抑制子是一类同源性较低的蛋白质家族,通过调控蔗糖转化酶抑制子的表达量可以抑制蔗糖转化酶活性。本课题组前期分离到甘薯转化酶抑制子IbINH全长ORF,本文对20种转化酶抑制子进行了生物信息学比较分析,可为深入研究IbINH基因功能提供参考。【方法】本研究以前期克隆获得的IbINH全长ORF推测的氨基酸序列和NCBI数据库中搜集到的其余19种植物INH氨基酸序列为分析数据来源,采用系列生物信息学分析软件对INH蛋白质的氨基酸组成及其理化性质、蛋白质亲疏水性、磷酸化位点、INH蛋白质的二级元件和含量、跨膜结构域和信号肽、蛋白质亚细胞定位、功能结构域和空间三维结构等进行预测和分析。【结果】根据预测和分析发现,IbINH同其他INH蛋白质一样均含有典型的PMEI-like超级家族功能结构域,二级结构组成和比例也比较相似,INH蛋白质定位在细胞质的可能性最大,但是各种INH的亲疏水性、稳定性以及跨膜结构域等也有差异。【结论】本文通过对INH蛋白质进行系列生物信息学分析和比较,为以后IbINH基因功能研究奠定了重要基础。
【Objective】Invertase is one of the key factors involved in the pathway of sugar signal regulation in plants.It plays very important role in the regulation of sugar metabolism,plant growth,disease and stress resistance.Invertase inhibitors are a family of proteins with low homology.The activity of invertase can be inhibited by regulating the expression of invertase inhibitors.【Method】In this study,IbINH and the INH amino acid sequences of 19 other plants collected from and NCBI databases were used as data sources.A series of bioinformatics analysis software were used to analyze the amino acid sequences.The chemical properties,hydrophilicity,phosphorylation sites,secondary components and contents of INH proteins,transmembrane domain and signal peptide,protein subcellular localization,functional domain and spatial structure were predicted and analyzed.【Result】According to the prediction and analysis,we found that IbINH had a typical PMEI-like superfamily functional domain which was the same as other INH proteins,and the secondary structure composition was also similar.The INH proteins might be located in the cytoplasm.However,the hydrophobicity,stability and transmembrane domain were not completely similar among the 20 INHs.【Conclusion】Our study laid an important foundation for the functional study of IbINH.
