摘要
目的探讨姜黄素抑制人胃癌SGC7901/ADR细胞增殖情况以及与多药耐药相关因子P170及GST-π表达规律的相关性。方法采用CCK-8检测不同浓度姜黄素对SGC7901/ADR细胞12 h、24 h、36 h的抑制情况;通过免疫细胞化学法分析各浓度梯度姜黄素作用36 h时P170及GST-π的表达差异;采用统计学方法进行分析。结果姜黄素作用于人胃癌SGC7901/ADR耐药细胞剂量越高、时间越长吸光度越低(P<0.05),各浓度梯度姜黄素作用36 h时GST-π阳性表达有降低趋势,但无统计学意义(P>0.05),而与P170阳性表达则无直接关联性。结论姜黄素可抑制人胃癌SGC7901/ADR耐药细胞的增殖,可能抑制GST-π的表达。
Objective To investigate the inhibition of Curcumin on the proliferation of human gastric cancer SGC7901/ADR cells and the relationship between Curcumin and the expression of multidrug resistance related factors P170 and GST-π.Methods Cell counting kit-8(CCK-8)was used to detect the inhibition of Curcumin at different concentrations on SGC7901/ADR cells for 12,24 and 36 hours;immunocytochemistry was applied to analyze the differences of P170 and GST-πexpression at 36 hours after Curcumin treatment at different concentrations;statistical method was used to analyze the results.Results The higher the dose of Curcumin on SGC7901/ADR cells and the longer the time,the lower the absorbance(P<0.05).After 36 hours of Curcumin treatment at different concentrations,the positive expression of GST-πdecreased,and the difference was not statistically significant(P>0.05),and had no direct correlation with positive expression of P170.Conclusion Curcumin can inhibit the proliferation of SGC7901/ADR cells,and may inhibit the expression of GST-π.
