摘要
目的:应用生物信息学方法挖掘食管鳞状细胞癌(ESCC)的相关基因,探讨其发病机制,为ESCC诊断和靶向治疗提供依据。方法:从GEO(Gene Expression Omnibus)数据库下载基因芯片数据集GSE100942和GSE17351,利用其分析工具GEO2R筛选ESCC的差异表达基因(DEGs)。运用数据库DAVID进行GO功能和KEGG通路富集分析,应用Cytoscape软件和STRING数据库构建相互作用网络和关键基因模块,挖掘ESCC靶基因。进一步通过ONCOMINE和UCSC数据库的临床组织样本证实靶基因与ESCC的关系。结果:共筛选出80个DEGs,富集分析显示差异基因在细胞分裂、胶原纤维组织、有丝分裂胞质分裂、纺锤体、中间体等方面存在显著富集。共挖掘出11个ESCC靶基因,经证实均在临床ESCC组织样本中存在显著高表达。其中NUSAP1、KIF20A、DEPDC1、TTK、CCNB1、NCAPG、AURKA这7个靶基因尚未在ESCC中有研究。结论:通过生物信息学挖掘出的与ESCC相关的7个新靶基因,可能是未来研究ESCC发病机制、临床诊断和治疗的重要靶点。
Objective:To provide targets for the pathogenesis,diagnosis and treatment of esophageal squamous cell carcinoma(ESCC),we excavated the related genes of ESCC by bioinformatics analysis.Methods:We downloaded gene datasets GSE100942 and GSE17351 from the Gene Expression Omnibus(GEO)database and identified the differentially expressed genes(DEGs)by GEO2 R analysis tools.Enrichment analysis of GO and KEGG pathways was constructed by using the DAVID database.The interaction network and key gene modules were performed by Cytoscape software and STRING database.Finally,target genes were excavated.The relationship between target genes and ESCC was further confirmed byclinical tissue samples from ONCOMINE and UCSC databases.Results:80 DEGs were selected,and the DEGs were mainly enriched in cell division,collagen fibril organization,mitotic cytokinesis,spindle and midbody.A total of 11 target genes were excavated.All of which were proved to be highly expressed in clinical ESCC tissue samples.Among them,the 7 target genes of NUSAP1,KIF20 A,DEPDC1,TTK,CCNB1,NCAPG and AURKA have not been studied in ESCC.Conclusion:Seven new target genes related to ESCC identified through bioinformatics may be important targets for future research on ESCC pathogenesis,clinical diagnosis and treatment.
