旋毛虫细胞外囊泡对TNBS诱导的小鼠实验性结肠炎的初步干预作用 被引量:3

Preliminary Prevention Effect of Trichinella spiralis Extracellular Vesicles on TNBS-Induced Experimental Colitis in Mice

摘要 为探讨旋毛虫细胞外囊泡(Trichinella spiralis extracellular vesicles,Ts-EVs)对2,4,6-三硝基苯磺酸(TNBS)诱导的小鼠实验性结肠炎的初步干预作用,通过超速离心法获得Ts-EVs,利用透射电子显微镜(TEM)对其进行鉴定。随后将30只Balb/c小鼠随机分为正常对照组、TNBS模型组和TNBS+Ts-EVs干预组。通过测定各组小鼠肠道疾病活动指数(DAI)、髓过氧化物酶(MPO)活性和结肠病理组织学变化等指标评价Ts-EVs对结肠炎的干预效果;进一步用ELISA检测外周血血清中细胞因子的表达情况。TEM结果显示,EVs具有膜结构,形态呈圆形或椭圆形,平均直径为30nm^150nm;评估Ts-EVs对TNBS诱导的小鼠结肠炎的干预效果表明,TNBS+Ts-EVs干预组小鼠的结肠炎临床症状有所缓解,炎症指标均明显降低;与TNBS模型组相比,TNBS+Ts-EVs干预组小鼠的Th1细胞因子(IL-1β、IFN-γ)和Th17细胞因子(IL-17A)表达水平均显著降低;而Treg细胞因子(IL-10、TGF-β)表达水平明显升高。研究结果表明Ts-EVs可有效干预TNBS诱导的小鼠实验性结肠炎,此干预作用可能与调节Th细胞免疫平衡有关。 To investigate the preliminary prevention effect of Trichinella spiralis extracellular vesicles(Ts-EVs)on 2,4,6-trinitrobenzene sulfonic acid(TNBS)-induced experimental colitis in mice,Ts-EVs were first obtained by ultracentrifugation and identified by transmission electron microscopy(TEM).30Balb/c mice were then randomly divided into control group,TNBS group and TNBS+Ts-EVs group.Intestinal disease activity index(DAI),body weight change,myeloperoxidase(MPO)activity and lais to histopathological lesions of colon were measured in each group to evaluate the prevention effect of Ts-EVs in colitis;ELISA was used to detect the expression of cytokines in serum.TEM results showed that EVs had a membrane structure and a circular or elliptical shape with an average diameter of 30nm-150nm.The evolution of the prevention effect of Ts-EVs on TNBS-induced colitis in mice showed that the clinical symptoms of colitis in the TNBS+Ts-EVs group were alleviated,and the inflammation indexes were significantly reduced.The ELISA results showed that the expression levels of Th1cytokines(IL-1β,IFN-γ)and Th17cytokine(IL-17A)were significantly lower in the TNBS+Ts-EVs group,while the expression levels of regulatory cytokines(IL-10,TGF-β)were significantly increased compared with the TNBS group.The results of this study indicated that Ts-EVs can effectively alleviate TNBS-induced colitis in mice.This prevention effect may be related with the regulation of the blance of Th cell immune resopones.
