Simultaneous determination of 14 constituents in Yuye Jinhua Qingre tablets by UPLC-ESI-MS/MS

摘要 目的建立UPLC-ESI-MS/MS法同时测定玉叶金花清热片中牛磺酸、京尼平苷酸、绿原酸、哈巴苷、山栀苷甲酯、3,5-O-二咖啡酰基奎宁酸、栀子苷、玉叶金花苷酸甲酯、β-蜕皮甾酮、穿心莲内酯、哈巴俄苷、脱水穿心莲内酯、胆红素、胆酸的含量。方法采用安捷伦ZORBAX SB C18柱(3.0 mm×50 mm,1.8μm),流动相为甲醇-0.1%乙酸(含0.02 mol·L^-1乙酸铵)溶液,梯度洗脱,体积流量0.3 mL·min^-1;质谱采用ESI正、负离子同时采集,多反应监测(MRM)模式扫描。结果14种成分在相应线性范围内与测定值线性关系良好(r>0.9985),平均加样回收率94.5%~101.5%(RSD<3.0%)。结论该方法简便、灵敏、高效,可用于玉叶金花清热片的质量控制。 Objcetive To simultanously determine the content of taurine,geniposidic acid,chlorogenic acid,harpagide,shanzhisidemethylester,3,5-O-dicaffeoylquinic acid,geniposide,mussaenoside,β-ecdysone,andrographolide,harpagoside,dehydroandrographolide,bilirubin,and cholic acid in Yuye Jinhua Qingre tablets by UPLC-ESI-MS/MS.Methods The chromatographic separation was achieved on an Agilent ZORBAX SB C18(3.0 mm×50 mm,1.8μm).The mobile phase consisted of methanol-0.1%acetic acid(containing 20 mmol·L^-1 ammonium acetate)at a flow rate of 0.3 mL·min^-1 with gradient elution,and the mass spectrum was scanned by ESI multiple reaction monitoring mode.Results Fourteen components showed good linearity within their own ranges(r>0.9985),whose average recoveries were at 94.5%-101.5%(RSD<3.0%).Conclusion This simple,sensitive,and effective method can be used for the quality control of Yuye Jinhua Qingre tablets.
