博落回叶肉组织原生质体的分离条件筛选 被引量:3

Isolation and Purification of Protoplasts from Mesophyll Tissues of Macleaya cordata

摘要 博落回(Macleaya cordata)是一种罂粟科博落回属的多年生草本植物,其体内的血根碱(sanguinarine,SAN)具有抗炎、调控肠道菌群和促进动物生长的作用,因此长期以此作为饲用抗生素的天然源替代品在全世界70个国家和地区广泛销售。随着全世界越来越多的国家限用甚至禁止饲用抗生素,博落回资源的需求逐年增加。因此,通过育种等方法来提高博落回中血根碱的含量具有十分重要的意义。而利用原生质体技术进行品种改良是一个快速高效的方法,其中制备原生质体是第一步。本研究以博落回无菌苗叶片为材料,进行了原生质体分离和纯化。在一定条件下,通过比较不同的酶解渗透压、温度、时间和pH值,得到最适甘露醇浓度为0.6 mol/L,最适温度为30℃,最适时间为6 h,最适酶液pH值为5.8,在该条件下的产量可达到6.03×10^5个/mL,活力为87.3%。该研究为未来博落回的种质资源创新提供了前期研究依据。 Macleaya cordata is a perennial herb of the genus Papaveraceae.It contains sanguinarine(SAN),which has anti-inflammatory properties,regulates intestinal flora and promotes animal growth.It has long been a natural source substitute for feed antibiotics,and widely used as a natural source substitute for feed antibiotics in 70 regions around the world.As more and more countries around the world restrict or even ban the use of feed antibiotics,the demand for resources is increasing year by year.Therefore,it is of great significance to improve the content of blood root alkali in M.cordata by methods such as breeding.The use of protoplast technology for variety improvement is a fast and efficient method,in which the preparation of protoplasts is the first step.In this study,protoplast isolation and purification were carried out using the leaves of sterile seedlings.Under certain conditions,by comparing different enzymatic osmotic pressure,temperature,time and pH value,the optimal osmotic pressure is 0.5 mol/L,the optimum temperature is 30℃,the optimum time is 6 h and the optimum pH value is 5.8.Under this condition,the yield can reach 6.03×10^5/mL,and the activity is 87.3%.This research provides a prelim-inary research basis for the future germplasm resources innovation.
