摘要
目的研究寨卡病毒(ZIKV)感染人神经母细胞瘤细胞(SH-SY5Y)所导致的细胞因子分泌的变化,探讨ZIKV影响神经系统功能紊乱的机制。方法建立ZIKV感染SH-SY5Y细胞模型,采用噻唑蓝(MTT)分析法检测细胞活力,采用悬浮芯片系统(Bio-Plex■200)Luminex多重检测技术同时定量检测24、48、72 h细胞培养上清液中细胞因子IL-1β、IL-2、IL-4、IL-5、IL-13、IL-18、IFN-γ、TNF-α、IL-12P70、GM-CSF的浓度。结果病毒感染组24、48、72 h细胞存活率均低于正常对照组(均P<0.01)。病毒感染组(24~72 h)分泌IL-1β、IL-5、IFN-γ、TNF-α、IL-12P70、GM-CSF的浓度水平均高于对照组(均P<0.01)。病毒感染组(48 h)分泌的IL-1β、IL-12P70、GM-CSF浓度水平均高于对照组(48 h),病毒感染组(72 h)分泌TNF-α高于对照组(72h)(P<0.05或P<0.01)。结论 ZIKV感染SH-SY5Y细胞后致其细胞存活率降低,并以时间依赖的方式分泌浓度水平相对较高的促炎性细胞因子。
Objective To observe changes in cytokine secretion induced by Zika virus(ZIKV)infection in human neuroblastoma SH-SY5 Y and explore the mechanism of ZIKV affecting nervous system dysfunction.Methods A model of SH-SY5 Y cells infected with ZIKV was established and the cell viability was detected by MTT assay.Cell culture supernatants were harvested at 24,48,and 72 hours.Concentrations of IL-1β,IL-2,IL-4,IL-5,IL-13,IL-18,IFN-γ,TNF-α,IL-12 P70,and GM-CSF were measured simultaneously and quantitatively using the Bio-Plex?200 system.Results The cell survival rates at 24,48,and 72 hours in the virus infection group were lower than those in the normal control group(all for P<0.01).The levels of IL-1β,IL-5,IFN-γ,TNF-α,IL-12 P70,and GM-CSF secreted by the virus infection group(24-72 h)were higher than those of the control group(all for P<0.01).The levels of IL-1β,IL-12 P70,and GM-CSF secreted by the virus infection group(48 h)were higher than those of the control group(48 h),and the level of TNF-αsecreted by the virus infection group(72 h)was higher than that of the control group(72 h)(P<0.05 or P<0.01).Conclusion SH-SY5 Y cells infected with ZIKV resulted in the decrease in cell viability and the secretion of relatively high concentration of pro-inflammatory cytokines in a time-dependent manner.
