摘要
目的探讨舒芬太尼联合右美托咪啶对脊髓损伤大鼠的治疗及可能机制。方法Allen’s法建立脊髓损伤模型,将SD大鼠分为假手术组(Sham组)、模型组(SCI组)和舒芬太尼与右美托咪啶联合治疗组(SD组);对各组大鼠脊髓运动功能(BBB)进行评分;应用HE染色检测各组大鼠脊髓的组织病理学变化;采用ELISA检测IL-10、IL-6及TNF-α水平变化;化学方法检测各组大鼠脊髓组织中MDA、CAT、SOD、GSH-Px等蛋白的表达情况;IHC检测各组脊髓组织中的HMGB1、TLR4表达;Western blot法检测HMGB1、TLR4、Bcl-2和Bax的表达水平。结果HE染色结果显示,两种药物的联合治疗能降低脊髓中炎症性中性粒细胞的浸润;与SCI组相比,SD组大鼠BBB评分显著升高,血清SOD、CAT、GSH-Px含量升高,MDA含量降低,脊髓组织中IL-10与Bcl-2的表达水平明显升高,TNF-α、IL-6、Bax、HMGB1及TLR的表达明显下降(P<0.05)。结论舒芬太尼与右美托咪啶联合应用能够改善大鼠的脊髓损伤,其机制可能与下调HMGB1/TLR4信号通路有关。
Objective To explore the therapeutic effect of sufentanil combined with dexmedetomidine on spinal cord injury in rats and its possible mechanism.Methods The spinal cord injury model was established by Allen’s method.SD rats were divided into sham group,SCI(spinal cord injury)group and SD(sufentanil and dexmedetomidine combined treatment)group.The spinal cord motor function(BBB)of each group was scored.The histopathological changes of spinal cord were detected by HE staining.The levels of IL-10,IL-6 and TNF-αwere detected by ELISA.The expression of MDA,CAT,SOD and GSH-Px in spinal cord tissues of rats in each group were detected by chemical methods;the expression of HMGB1 and TLR4 in spinal cord tissues of rats in each group was detected by IHC;the expression levels of HMGB1,TLR4,Bcl-2 and Bax were detected by western blot.Results The results of HE staining showed that the combination of the two drugs could reduce the infiltration of inflammatory neutrophils in the spinal cord;compared with SCI group,the BBB score of rats in SD group was significantly higher,the content of serum SOD,CAT,GSH-Px was higher,the content of MDA was lower,the expression of IL-10 and Bcl-2 in the spinal cord tissue was significantly higher,the expression of TNF-α,IL-6,Bax,HMGB1 and TLR were significantly lower(P<0.05).Conclusion Sufentanil combined with dexmedetomidine can improve spinal cord injury in rats,and its mechanism may be related to down regulating HMGB1/TLR4 Signal pathway.
