摘要
为研究山西地方猪种的肉质相关基因表达量,选取30 kg左右的山西黑猪和晋阳白猪各30头,按品种分为山西黑猪组和晋阳白猪组,每组5个重复,每个重复6头猪,进行驱虫和健胃后,开始正式试验,饲喂相同的饲粮,猪瘟、口蹄疫、蓝耳病等免疫程序按常规进行。体重达110 kg左右时屠宰,从背最长肌取样,提取基因组,进行PCR扩增,检测钙蛋白酶抑制蛋白(CAST)、猪心型脂肪酸结合蛋白(H-FABP)、肌细胞生成素(MyoG)、ATP合成酶(ATP5B)、脂蛋白脂肪酶(LPL)、磷酸果糖激酶(PFK)、己糖激酶同工酶(HK)等7个肉质相关基因的相对表达量。结果表明,山西黑猪HK、MyoG基因的相对表达量极显著高于晋阳白猪(P<0.01);山西黑猪H-FABP、CAST基因的相对表达量显著高于晋阳白猪(P<0.05);山西黑猪ATP5B、PFK、LPL基因的相对表达量与晋阳白猪差异不显著(P>0.05)。
In order to study the expression of meat-related genes in Shanxi local pig breeds,30 Shanxi black pigs and 30 Jinyang white pigs of about 30 kg were selected and divided into Shanxi black pig group and Jinyang white pig group.There were 5 repetitions in each group,6 pigs in each repetitions.After deworming and stomach strengthening were carried out,and the formal experiment was started,feed the same basic diet.The immune procedures of classical swine fever,foot-and-mouth disease and blue ear disease were carried out according to the routine.Slaughtering when the weight was about 110 kg.The genome was extracted from the longissimus dorsi muscle and amplified for PCR.The amplification and dissolution curves were generated automatically.The relative expression levels of seven meat related genes,such as calpain inhibitory protein(CAST),porcine heart fatty acid binding protein(H-FABP),MyoGenin(MyoG),ATP5B,LPL,PFK and HK,were detected.The results showed that the relative expression of HK,MyoG genes in Shanxi black pigs was significantly higher than that of Jinyang white pigs(P<0.01);the relative expression of H-FABP,CAST genes in Shanxi black pigs were significantly higher than that of Jinyang white pigs(P<0.05);the expression levels of ATP5B,PFK and LPL genes in Shanxi black pigs were not significantly different from those in Jinyang white pigs(P>0.05).
