摘要
目的研究小鼠子宫中经典瞬时感受器电位通道6(transient receptor potential canonical 6,TRPC6)离子通道在离体子宫收缩中的作用。方法分离野生型(WT)或TRPC6-KO小鼠子宫,使用等张张力测试系统观察TRPC6通道抑制剂Pyr10和Larixyl对离体小鼠子宫收缩的影响;分离WT或TRPC6-KO小鼠子宫平滑肌细胞,使用钙成像技术观察催产素引起的细胞内钙离子浓度的变化。结果Pyr10和Larixyl能明显抑制催产素引起的子宫收缩;催产素引起的TRPC6-KO小鼠子宫的收缩持续时间明显较WT的长;催产素引起TRPC6-KO的子宫平滑肌细胞钙离子浓度升高后减退速度较WT缓慢。结论TRPC6通道介导的钙离子内流参与了小鼠子宫的收缩,并且催产素引起的小鼠子宫收缩随时间的衰减可能与TRPC6通道电流的衰减有关。
Aim To investigate the role of TRPC6 channel in the spontaneous and oxytocin(OT)-induced contractions of mouse uterus in vitro.Methods The uteruses of wildtype(WT)and TRPC6-KO mice were isolated,and the effects of TRPC6 channel inhibitors on uterine contraction were monitored by isometric tension test system in vitro.Smooth muscle cells from tested uterus were isolated by collagenase P,and the intracellular calcium signals were recorded by calcium imaging.Results TRPC6 channel inhibitors Pyr10 and Larixyl significantly suppressed OT-induced uterine contraction in vitro.Furthermore,the attenuation of OT-induced contraction was slower in TRPC6-KO uterus.The decay rate of OT induced calcium signal in TRPC6-KO uterine smooth muscle cells was slower than WT.Conclusions Calcium influx mediated by TRPC6 channel contributes to OT-induced mouse uterine contraction,and the attenuation of the contraction may relate to the decay of activation of TRPC6 current.
