摘要
本试验以切花小菊品种瑞白,紫丹特的茎尖为外植体,探究切花小菊茎尖离体培养及一次成苗培养基。结果表明:适合瑞白外植体的最佳灭菌时间为8 min,培养基为1/2MS+1.0 mg/L6-BA+0.3 mg/LNAA;适合紫丹特外植体的最佳灭菌时间为6 min,培养基为1/2MS+1.0 mg/L6-BA+0.3 mg/LNAA;适合2个品种切花小菊组培苗移栽的栽培基质草炭与珍珠岩之比为3:1,且成活率分别为96%,98%。
In this study, the stem tips of chrysanthemum varieties Rebekah and Purple Dent were used as explants to explore the culture medium of chrysanthemum stem tips in vitro and seedling culture. The results showed that the optimal sterilization time for Ruibai explants was 8 min and the medium was 1/2 ms +1.0 mg/L6-ba +0.3 mg/LIBA. The optimal sterilization time for Zidantte explants was 6 min, and the medium was 1/2 ms +1.0 mg/L6-ba +0.3 mg/LIBA. The substrates suitable for the transplantation of tissue culture seedlings of the two varieties of cut chrysanthemum were peat: perlite 3:1, and the survival rates were 96% and 98% respectively.
