摘要
目的比较microRNA-25在结肠癌患者与健康人群血浆中的表达差异,探讨microRNA-25在诊断结肠癌中的价值。方法选取结肠癌患者50例作为试验组,另匹配一般资料相似的健康体检者作为健康组,收集2组患者的血浆标本,采用实时荧光定量聚合酶链反应(qRT-PCR)检测血浆中microRNA-25的表达水平并分析其在不同临床病理资料间的差异。运用受试者工作曲线(ROC)计算microRNA-25在结肠癌中的诊断价值。结果microRNA-25血浆中的相对表达量显著低于健康组,差异有统计学意义(P=0.001);结肠癌患者血浆中的microRNA-25与年龄、性别、肿瘤位置以及CEA水平无相关性,而与临床病理分级(TNM分级)、淋巴结转移与否呈正相关(P<0.05)。microRNA-25作为结肠癌的生物标志物(AUC)曲线下的面积为0.906,以2.06为相对表达量的节点,特异度为66.7%和灵敏度为96.2%,尤登指数为0.85。结论microRNA-25在结肠癌患者血浆中呈高表达水平,推测microRNA-25可能结肠癌的发生发展过程,有望成为预测结肠癌的检测指标之一。
Objective To investigate the expression difference and clinical significance of serum microRNA-25 between the patients with colorectal cancer and healthy people.Methods Fifty blood specimens were extracted from the patients with colorectal cancer(experimental group),at the same time,the thirty blood specimens were extracted from healthy people(control group).The expression levels of microRNA-25 were detected by qRT-PCR method,and the differences of expression levels of microRNA-25 in clinical patholoxial parameters were analyzed.The application value of microrna-25 in diagnosis of colon cancer was evaluated by means of the subject’s work curve(ROC).Results The expression levels of microRNA-25 in experimental group(4.16±0.74)were significantly lower than those(1.04±0.38)in control group(P<0.01).The expression levels of microRNA-25 in experimental group were not correlated with patient’s age and gender,tumor position and CEA levels,however,which were positively correlated with the TNM stage and the lymph node metastasis(P<0.05).Moreover the area under AUC of microRNA-25 as biological marker of colon carcinoma was 0.906,taking 2.06 as the nodal point of relative expression levels,the specificity and sensitivity were 66.7%and 96.2%,respectively,with Youden’s index being 0.85.Conclusion The microRNA-25 is highly expressed in patients with colorectal cancer,which may be involved in the pathogenesis and development of colorectal cancer,thus it might be one of biomarkers of predicting colorectal cancer.
