摘要
目的:建立超高效液相色谱(UPLC)法同时测定五味沙棘散中7个成分含量的方法。方法:色谱柱为AQUITY UPLC BEH C18柱(100 mm×2.1 mm,1.8μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,流速为0.2 ml·min-1,检测波长为250 nm、370 nm,柱温为30℃,进样量为1μl。采用统计分析软件对含量测定结果进行聚类分析与主成分分析。结果:栀子苷、甘草苷、槲皮素、异鼠李素、甘草酸、木香烃内酯和去氢木香内酯检测质量浓度线性范围分别为49.53~445.80μg·ml-1、5.86~52.74μg·ml-1、8.35~75.12μg·ml-1、11.80~106.20μg·ml-1、16.74~150.70μg·ml-1、41.73~375.60μg·ml-1、60.23~542.20μg·ml-1(r≥0.9992);平均加样回收率分别为98.6%,98.9%,98.8%,99.0%,99.0%,98.9%,99.3%,RSD分别为0.3%,0.2%,0.1%,0.4%,0.3%,0.3%,0.2%(n=9)。同一厂家产品质量具有较高的稳定性,不同厂家样品存在一定差异。结论:本法简便、准确、重复性好,可用于五味沙棘散的质量控制,提示厂家在生产过程中要加强对沙棘膏和木香质量的控制,为五味沙棘散质量提高提供参考。
Objective:To establish a UPLC method for the simultaneous determination of 7 constituents in Wuwei Shaji powder.Methods:An AQUITY UPLC BEH C18 column(100 mm×2.1 mm,1.8μm)was used with the column temperature at 30°C.The injection size was 1μl.The mobile phase was acetonitrile(A)-0.1%phosphoric acid solution(B)with gradient elution at the flow rate of 0.2 ml·min-1.The detection wavelengths were 250 nm and 370 nm.The principal component analysis and cluster analysis were conducted for the results of content determination by statistical software.Results:The linear range was 49.53-445.80μg·ml-1 for geniposide,5.86-52.74μg·ml-1 for liquiritin,8.35-75.12μg·ml-1 for quercitrin,11.80-106.20μg·ml-1 for isorhamnetin,16.74-150.70μg·ml-1 for ammonium glycyrrhizate,41.73-375.60μg·ml-1 for costunolide and 60.23-542.20μg·ml-1 for dehydrocostuslactone(r≥0.9992).The average recovery was 98.6%,98.9%,98.8%,99.0%,99.0%,98.9%and 99.3%,and RSDs were 0.3%,0.2%,0.1%,0.4%,0.3%,0.3%and 0.2%(n=9),respectively.The quality of the samples from the same manufacturer had high stability,while there were some differences among 15 batches of samples from different manufacturers.Conclusion:The method is simple and accurate with good repeatability,which can be used to control the quality of Wuwei Shaji powder.It is suggested that the quality control of Hippophae Fructus and Aucklandiae Radix should be strengthened in the process of production to provide support for the quality improvement of Wuwei Shaji powder.
