摘要
目的检测肉豆蔻酰化的富含丙氨酸的C激酶底物(MARCKS)、人类表皮生长因子受体2(HER2)在胃癌组织中的表达水平,分析MARCKS、HER2表达与胃癌患者临床病理参数和预后的关系,并探讨两者在胃癌组织中表达的相关性。方法收集我院2013年1月至2014年12月诊疗的110例胃癌患者的胃癌组织和癌旁组织,应用免疫组化(IHC)检测胃癌和癌旁组织中MARCKS和HER2的表达情况;分析MARCKS、HER2的表达水平与胃癌患者临床病理参数;Logrank检验分析MARCKS、HER2的表达水平与胃癌患者预后的关系;Pearson线性相关分析MARCKS与HER2在胃癌组织中表达的相关性。结果IHC检测MARCKS、HER2在胃癌组织中的阳性表达率均高于在癌旁组织中的阳性表达率(P<0.05)。胃癌组织中MARCKS的阳性表达与患者分化程度、淋巴结转移和TNM分期相关,HER2的阳性表达与淋巴结转移和TNM分期相关(P<0.05)。MARCKS、HER2阳性表达组胃癌患者生存率低于MARCKS、HER2阴性表达组患者(P<0.05)。胃癌组织中MARCKS与HER2的表达呈正相关(P<0.05)。结论MARCKS、HER2阳性表达与胃癌患者的不良病理参数和预后相关,两者可能发挥协同作用共同促进胃癌的进展,是潜在的不良预后标志物和治疗靶标。
Objective To detect myristoylated alanine-rich C-kinase substrate(MARCKS),human epidermal growth factor receptor 2(HER2)expression levels in gastric cancer tissues,so to analyze the relationship between the expressions of MARCKS and HER2 and the clinicopathological parameters and prognosis of gastric cancer,and further to explore the correlation between the expressions of MARCKS and HER2 in gastric cancer.Methods The gastric cancer tissues and adjacent tissues of 110 patients with gastric cancer diagnosed and treated in our hospital from January,2013 to December,2014 were collected for the study.Immunohistochemistry(IHC)was used to detect the expressions of MARCKS and HER2 in gastric cancer and adjacent tissues.The expression levels of MARCKS and HER2,as well as clinicopathological parameters of gastric cancer patient were analyzed.Logrank test was used to analyze the relationship between the expression levels of MARCKS,HER2 and the prognosis of gastric cancer patients.Pearson linear correlation analysis was used to detect the correlation between MARCKS and HER2 expressions in gastric cancer tissues.Results The positive expression rates of MARCKS and HER2 in gastric cancer tissues detected by IHC were higher than those in adjacent tissues(P<0.05).The positive expression of MARCKS in gastric cancer tissues was correlated with differentiation degree,lymph node metastasis and TNM stage(P<0.05).The positive expression of HER2 in gastric cancer tissues was correlated with lymph node metastasis and TNM stage(P<0.05).The survival rate of patients with gastric cancer in MARCKS and HER2 positive expression group was lower than that in patients with MARCKS and HER2 negative expression group(P<0.05).There was a positive correlation between MARCKS and HER2 expressions in gastric cancer tissues(P<0.05).Conclusion The positive expressions of MARCKS and HER2 are related to the adverse pathological parameters and prognosis of patients with gastric cancer.These proteins may play a synergistic role to promote the progression of gastric cancer,and they can serve as potential markers of poor prognosis and used as therapeutic targets.
