氧化铅悬浊液灌注造影在皮瓣微血管研究中的应用

摘要 目的探讨氧化铅悬浊液在皮瓣微血管造影中的方法和效果。方法取8周龄无特定病原体(SPF)级Wistar大鼠18只(购自北京维通利华实验动物技术有限公司),随机分为6组。颈动脉插管后通过生物信号采集与处理系统测量大鼠收缩压,以生理盐水配制25%、30%、35%、40%、45%和50%共6组不同浓度的氧化铅悬浊液,分别对各组大鼠进行血管灌注并根据波意尔定律计算灌注压强。灌注成功后将大鼠标本冷冻24 h进行全身X线摄影,随后解剖腹部皮瓣进行大体观察及X线微血管摄影,并对收集整理的数据进行t检验。结果氧化铅悬浊液灌注压强均高于大鼠收缩压,灌注压强/大鼠收缩压随氧化铅浓度的增加显著升高(25%,1.34±0.11;30%,1.86±0.33;35%,2.16±0.21;40%,3.37±0.93;45%,7.29±0.59;50%,13.65±0.87),组间差异有统计学意义(t=3.520、3.190、1.950、14.860、16.350,P<0.05)。18只大鼠均灌注成功,浓度为35%和40%的各组皮瓣微血管显影清晰,效果最好。结论氧化铅悬浊液灌注造影操作简单、安全,对皮瓣血管有较好的显影效果,可作为传统方法的替代和补充。浓度应配制在35%~40%为宜,且需要在较短时间内迅速匀速推注灌注液。 Objective To explore the method and effect of lead oxide suspension perfusion in skin flap microvessels angiography.Methods There were 188-week-old specific pathogen free(SPF)Wistar rats by grouping randomly in this study.The systolic pressure of rats was measured by biological signal acquisition and processing system after carotid artery intubation.Six groups of lead oxide suspension with different concentrations of 25%,30%,35%,40%,45%and 50%were prepared.Perfusion pressure was calculated according to Boyle’s law.After successful perfusion,the rats were frozen for 24 h for X-ray radiography.Then the abdominal flap was dissected for gross observation and microangiography.The t test was conducted to analyze the collated data.Results The perfusion pressure was higher than the systolic pressure and the ratio of perfusion pressure to the systolic pressure increased with the increase of lead oxide concentration(25%,1.34±0.11;30%,1.86±0.33;35%,2.16±0.21;40%,3.37±0.93;45%,7.29±0.59;50%,13.65±0.87),with statistical significance(t=3.520,3.190,1.950,14.860 and 16.350,P<0.05).All the rats were successfully perfused and the perfusion effects in 35%and 40%groups were best.Conclusion Lead oxide suspension perfusion is convenient and safe.It can be an alternative to the traditional methods.The concentration should be prepared between 35%to 40%,and the infusion should be performed in a short time at a rapid and uniform speed.
