摘要
目的:研究金刚藤胶囊抗慢性盆腔炎大鼠盆腔粘连的作用及其药理机制。方法:选择未交配雌性大鼠,随机分为假手术对照组、模型对照组、妇科千金片0. 15 g/kg组、金刚藤胶囊1. 2、2. 4、4. 8 g/kg组。除假手术对照组外,各组大鼠均采用机械和混合细菌液感染双重损伤法制作盆腔粘连模型。造模后,除假手术对照组、模型对照组外,各组大鼠灌胃给予相应药物连续20 d。末次药后,采用Verco评价标准对大鼠盆腔粘连进行评分,取子宫组织进行形态观察和HE染色进行病理观察,ELISA法测定血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、局部粘连组织的转化生长因子-β(TGF-β)、基质金属蛋白酶-9(MMP-9)含量以及组织型纤溶酶原激活剂(t PA)、纤溶酶原激活剂抑制因子-1(PAI-1)活性。荧光定量PCR法检测大鼠子宫组织TGF-β、MMP-9、t PA和PAI-1mRNA的表达。结果:与假手术对照组比较,模型对照组大鼠子宫盆腔粘连评分和组织病理病变程度增加,血清TNF-α、IL-β含量明显升高,子宫组织的TGF-β含量、PAI-1活性增高,mRNA相对表达量上调,而MMP-9含量、t PA活性明显降低,其mRNA表达明显下调(P <0. 05或P <0. 01)。与模型对照组比较,金刚藤胶囊2. 4、4. 8 g/kg组和妇科千金片组使大鼠子宫盆腔粘连评分和组织病理病变程度降低,血清TNF-α、IL-β含量明显下降,子宫组织的TGF-β含量和PAI-1活性明显降低,其mRNA相对表达量均明显下调,而MMP-9含量、t PA活性明显升高,其mRNA表达均明显上调(P <0. 05或P <0. 01)。结论:金刚藤胶囊能改善慢性盆腔炎大鼠的子宫炎症及炎性粘连,其作用与调节粘连形成因子和粘连蛋白溶解因子的表达有关。
Objective: To study the effect of Jingangteng capsule on chronic pelvic inflammation in rats with pelvic adhesion and its mechanism.Methods: Unmated female rats were randomly divided into four groups,the sham operation group,the model group,Fukeqianjin tablet( 0.15 g/kg) group,Jingangteng capsule groups( 1. 2,2. 4,4. 8 g/kg). Except for the sham operation group,the pelvic adhesion model was established by mechanical damage and mixed bacterial solution infection. After modeling,corresponding drugs were orally given to rats,except for the sham operation group and the model group. After the last administration,the pelvic adhesions of rats were scored by Verco evaluation standard. Uterine tissues were taken for morphological observation and HE staining was used for pathological observation. The serum tumor necrosis factor-α( TNF-α),Interleukin-1β( IL-1β),the content of transforming growth factor-β( TGF-β),matrix metalloproteinases-9( MMP-9),the activities of tissue-type plasminogen activator( t PA) and plasminogen activator inhibitor-1( PAI-1) in the local adhesions tissue were detected by ELISA. The mRNA expression of TGF-β,MMP-9,t PA and PAI-1 in rat uterus was detected by real-time PCR. Results: Compared with the sham operation group,the pelvic adhesion scores and histopathological lesions of uterine in the model group were increased,the levels of serum TNF-α and IL-β,TGF-β and PAI-1 activity and their mRNA expression in uterine tissues were significantly increased,while MMP-9 content and t PA activity and their mRNA expression were decreased. Compared with the model group,the pelvic adhesion scores and histopathological lesions of uterine were decreased in Jingangteng capsule groups( 2. 4、4. 8 g/kg) and Fukeqianjin tablets group,the levels of serum TNF-α and IL-β,TGF-β and PAI-1 activity and their mRNA expression in uterine tissues were significantly decreased,while MMP-9 content and t PA activity and their mRNA expression were increased. Conclusion: Jingangteng capsule can improve the uterine inflammation and inflammatory adhesion induced by chronic pelvic inflammation in model rats. Its mechanism may be related to the regulation of adhesion forming factor and adhesion dissolving factor.
