电针超前镇痛对脊髓背角神经元及星形胶质细胞活化的影响 被引量:13

Effect of electroacupuncture preemptive analgesia on activation of spinal dorsal horn neurons and astrocytes

摘要 目的观察电针的超前镇痛效应及对脊髓背角神经元及星形胶质细胞活化的影响。方法将80只SD大鼠随机分为正常组、模型组、预电针组、后电针组,每组20只。模型组、预电针组、后电针组采用4%甲醛右侧足底注射方法构建急性炎性疼痛模型,正常组右侧足底注射等量生理盐水,电针取同侧足三里、阳陵泉穴。预电针组先进行电针刺激30 min后再造模,造模后继续捆绑30 min;后电针组前30 min仅捆绑不进行电针刺激,造模后立即行电针刺激30 min;正常组和模型组先捆绑30 min后造模,再捆绑30 min。观察各组大鼠行为学变化(缩足次数和舔足时间),采用免疫荧光染色及Western blot法观察各组大鼠脊髓背角神经元、星形胶质细胞活化情况及c-fos和GAFP表达情况,ELISA法检测各组大鼠脊髓中干扰素-γ(IFN-γ)、P物质含量。结果与正常组相比,模型组大鼠缩足次数明显增加(P<0.05),舔足时间明显延长(P<0.05);与模型组相比,预电针组、后电针组大鼠缩足次数均明显减少(P均<0.05),舔足时间均明显缩短(P均<0.05),且预电针组效果优于后电针组(P均<0.05)。与正常组相比,模型组大鼠脊髓背角神经元、星形胶质细胞的活化明显增强,脊髓中c-fos和GAFP的表达水平、IFN-γ及P物质含量均明显增高(P均<0.05);与模型组相比,预电针组、后电针组大鼠脊髓背角神经元、星形胶质细胞的活化明显减弱,脊髓中c-fos和GAFP的表达水平、IFN-γ及P物质含量均明显降低(P均<0.05),且预电针组改善情况明显优于后电针组(P均<0.05)。结论电针预刺激可通过抑制脊髓背角神经元、星形胶质细胞的活化,减弱神经炎症以及疼痛放大,从而发挥超前镇痛效应。 Objective It is to observe the preemptive analgesic effect of electroacupuncture and its effect on the activation of spinal dorsal horn neurons and astrocytes.Methods Eighty SD rats were randomly divided into normal group,model group,pre-electroacupuncture group and post-electroacupuncture group,with 20 rats in each group.The model group,pre-electroacupuncture group,and post-electroacupuncture group were given the injection of 4%formaldehyde on the right foot plantar to establish acute inflammatory pain models.The normal group was injected with the same amount of normal saline on the right plantar foot.The electroacupuncture was performed in Zusanli and Yanglingquan acupoints at the same side.The pre-electroacupuncture group was treated with electroacupuncture for 30 minutes,and then the models were established.After the models were established,the binding was continued for 30 minutes.The post-electroacupuncture group was bundled without electroacupuncture stimulation at 30 min before modeling and was treated with electroacupuncture for 30 minutes immediately after modeling.The normal group and model group was bundled for 30 minutes and then were modeled,and then were bundled for 30 minutes.The behavioral changes(foot shrinkage and foot licking time)of each group were observed.Immunofluorescence staining and Western blot were used to observe the activation of neurons,astrocytes,c-fos and GAFP in the spinal dorsal horn of each group.The contents of interferon-γ(IFN-γ)and substance P in the spinal cord of rats in each group were measured by ELISA.Results Compared with the normal group,the number of foot reduction in the model group was significantly increased(P<0.05),and the licking time was significantly prolonged(P<0.05);compared with the model group,the number of foot reduction was significantly decreased(P<0.05),and the licking time was significantly shortened in the pre-electroacupuncture group and post-electroacupuncture group(all P<0.05),and the effect of the pre-electroacupuncture group was better than that of the post-electroacupuncture group(P<0.05).Compared with the normal group,the activation of neurons and astrocytes in the dorsal horn of the spinal cord of the model group was significantly enhanced,the expression levels of c-fos and GAFP,the content of IFN-γand substance P in the spinal cord were significantly increased(P<0.05);compared with the model group,the activation of dorsal horn neurons and astrocytes in the spinal cord of the pre-electroacupuncture group and the post-electroacupuncture group was significantly weakened,and the expression levels of c-fos and GAFP and the content of IFN-γand substance P in the spinal cord was significantly reduced(all P<0.05),and the improvement of the pre-electroacupuncture group was significantly better than that of the post-electroacupuncture group(P<0.05).Conclusion Electroacupuncture pre-stimulation can play a preemptive analgesic effect by inhibiting the activation of spinal dorsal horn neurons and astrocytes,weakening neuroinflammation and pain amplification.
