摘要
以绿头鸭(Anas platyrhynchos)为样本,采用巣式聚合酶链式反应(巣式PCR)方法克隆出Toll样受体7(TLR7)开放式阅读框架(ORF)的全长,对序列进行生物信息学分析,采集绿头鸭的心、肝、脾、肺、肾、肠6个部位试样进行荧光定量分析,探索绿头鸭TLR7基因的结构和它在不同器官的分布。结果表明:绿头鸭TLR7的ORF全长3144 bp,共编码1047个氨基酸。蛋白质结构预测工具(SMART)程序预测显示,TLR7具有典型的胞内功能区、跨膜区、胞外区。同源性分析表明,绿头鸭TLR7与鸟类同源性最高,与哺乳类、两栖类、鱼类形成明显分支,具有高度的种属特异性。荧光定量显示,TLR7在测试部位都有分布,在脾中最高,肝、肠次之,肺、心最低。
In order to explore the structure of Toll-like receptor 7(TLR7)gene and its distribution in different organs of Anas platyrhynchos,the full length of the Open Reading Frame(ORF)of TLR7 was cloned by nested PCR,the sequence was analyzed by bioinformatics,and fluorescence quantitative analysis was carried out on six parts of heart,liver,spleen,lung,kidney and intestine of collecting A.platyrhynchos.The ORF of TLR7 of A.platyrhynchos was 3144 bp in length,encoding 1047 amino acids.Protein structure prediction tool(SMART)program predicted that it had a typical intracellular Toll/interleukin-1 receptor domain,a transmembrane domain,an extracellular domain.The homology analysis showed that the TLR7 of the A.platyrhynchos had the highest homology with the birds,and formed obvious branches with mammals,amphibians and fishes,which had a high species specificity.Quantitative real-time PCR showed that TLR7 was distributed at the test site,the highest in the spleen,followed by the liver and intestine,and the lung and heart were the lowest.
