3种规格菊花有效成分与重金属元素的质量评价研究 被引量:4

Study on the Quality Assessment of Three Kinds of Juhua(Chrysanthemum)by Effective Components and Heavy Metal Elements

摘要 目的:以绿原酸(C)、木犀草苷(L)、3,5-O-二咖啡酰基奎宁酸(D)3种有效成分的含量和铅(Pb)、镉(Cd)、砷(As)、汞(Hg)、铜(Cu)5种重金属元素的含量综合评价3种菊花(杭菊、贡菊、胎菊)的质量,并研究二者之间的相关性。方法:从浙江、安徽收集33份杭菊、16份贡菊、10份胎菊样品,建立测定这3种有效成分含量的HPLC法和5种重金属含量的ICP-MS法,测定其含量,运用单因素方差分析法、主成分分析法比较3种规格菊花的差异,并结合多元相关性分析法研究这3种有效成分与5种重金属之间的相关性。结果:杭菊与贡菊的C、D含量差异,杭菊与胎菊之间的C含量差异,贡菊和胎菊之间的D含量差异均有统计学意义(P<0.05)。主成分分析的综合得分值贡菊(0.006,8,n=16)>胎菊(0.006,0,n=10)>杭菊(0.005,1,n=33)。杭菊和贡菊的Pb、Cu、Hg含量差异,杭菊和胎菊之间的Pb、Cu含量差异均有统计学意义(P<0.05)。主成分分析的综合得分值杭菊(1.747,9,n=33)>贡菊(0.516,6,n=16)>胎菊(0.196,0,n=10)。多元相关性分析表明,L与Pb成负相关,表达式为L=-0.264Pb+0.003;D与Pb成负相关、与Hg成正相关,表达式为D=-0.260Pb+0.394Hg+0.009;其余之间均无相关性。结论:建立测定3种规格菊花中3种有效成分含量的HPLC法和5种重金属元素含量的ICP-MS方法准确、可靠、重现性好。以药效的角度而言,贡菊的质量最优,其次是胎菊,杭菊最差。以安全性的角度考虑,胎菊的安全性最高,其次是贡菊,杭菊最低。二者存在一定的相关性。 Objective:To comprehensively assess the quality of three kinds of Juhua(Chrysanthemum)[Hangju(Chrysanthemum morifolium),Gongju(Chrysanthemum)and Taiju(Tire Chrysanthemum)]based on chlorogenic acid(C),luteoloside(L)and 3,5-Dicaffeoyl quinic acid(D)content of three active components and plumbum(Pb),cadmium(Cd),arsenic(As),mercury(Hg)and copper(Cu)content of five kinds of heavy metal elements,and to study the correlation both of them.Methods:Totally 33 copies of Hangju(Chrysanthemum morifolium),16 copies of Gongju(Chrysanthemum)and 10 copies of Taiju(Tire Chrysanthemum)samples were collected from Zhejiang and Anhui.ICP-MS and HPLC methods was developed for determination of the three active ingredient content and five kinds of heavy metal content,respectively,and its content was determined.The three kinds of chrysanthemum were compared by the difference of single factor variance analysis method and principal component analysis method.Multivariate correlation analysis was applied to explore the correlation between the law research of the three active ingredients and five kinds of heavy metals.Results:There are statistically significant difference(P<0.05)on the content of C and D between Hangju(Chrysanthemum morifolium)and Gongju(Chrysanthemum),the content of C between Hangju(Chrysanthemum morifolium)and Taiju(Tire Chrysanthemu),and the content of D between Gongju(Chrysanthemum)and Taiju(Tire Chrysanthemu).The comprehensive score of principal component analysis show that Gongju(Chrysanthemum)(0.006,8,n=16)>Taiju(Tire Chrysanthemu)(0.006,0,n=10)>Hangju(Chrysanthemum morifolium)(0.005,1,n=33).There are statistically significant difference(P<0.05)on the content of Pb,Cu and Hg between Hangju(Chrysanthemum morifolium)and Gongju(Chrysanthemum),and the content of Pb and Cu between Hangju(Chrysanthemum morifolium)and Taiju(Tire Chrysanthemu)is the same as it.The comprehensive score of principal component analysis show that Hangju(Chrysanthemum morifolium)(1.747,9,n=33)>Gongju(Chrysanthemum)(0.516,6,n=16)>Taiju(Tire Chrysanthemu)(0.196,0,n=10).Multivariate correlation analysis showed that L was negatively correlated with Pb,and the expression was L=-0.264Pb+0.003.D is negatively correlated with Pb and positively correlated with Hg.The expression is D=-0.260Pb+0.394Hg+0.009,and there was no correlation among the others.Conclusions:The developed HPLC method for determining the content of three effective components and ICP-MS method for determining the content of fivekinds of heavy metal content in three kinds of chrysanthemum is accurate,reliable and reproducible.The medicine effect of point,the best quality is Gongju(Chrysanthemum),Taiju(Tire Chrysanthemum)secondly,and Hangju(Chrysanthemum morifolium)is the worst.To consider the safety of point,the highest safety Taiju(Tire Chrysanthemum),Gongju(Chrysanthemum)is the second,and Hangju(Chrysanthemum morifolium)is lowest.There is a certain correlation both of them.
