枸杞多糖抑制小鼠肝纤维化组织中α-SMA和Ⅰ型胶原蛋白的表达 被引量:4

Inhibition on expression of alpha-SMA and typeⅠcollagen in liver fibrosis tissue of mice by Lycium barbarum polysaccharide

摘要 目的探讨枸杞多糖(LBP)对小鼠胆总管结扎术后引起的肝纤维化组织中α-平滑肌肌动蛋白(α-SMA)和Ⅰ型胶原蛋白(ColⅠ)表达的影响。方法选取体重为(0.32±0.28)kg的健康、雄性小鼠30只,采用完全随机方法分成对照组、模型组及LBP干预组,每组10只。模型组:小鼠通过结扎胆总管后诱导肝纤维化损伤。LBP干预组:小鼠在结扎胆总管后第21天口服LBP 30μg/g,每天1次,共用15天。对照组:小鼠暴露腹腔、分离、置线,不对胆总管做其他操作,闭合腹腔后第21天口服等剂量的0.90%生理盐水,每天1次,共用15天。制备3组的肝组织标本,采用苏木素-伊红染色法、天狼猩红染色法分析其肝脏组织的病理学变化;免疫印迹法测定3组肝组织中α-SMA和ColⅠ的含量。结果模型组小鼠肝小叶结构被破坏,肝组织有面积不等的坏死,汇管区发现胆总管数量增加和纤维组织增生,表明采用胆总管结扎术可以成功地诱导肝纤维化损伤,肝组织中α-SMA和ColⅠ的含量(0.877±0.015,1.102±0.063)增加,与对照组(0.286±0.005,0.765±0.017)比较,差异均有统计学意义(均P<0.05)。LBP干预组小鼠肝组织坏死减少、胆总管数目减少和纤维组织增生减轻,肝组织中α-SMA和ColⅠ的含量(0.302±0.011,0.456±0.010)减少,与模型组比较,差异有统计学意义(P<0.05)。结论LBP可抑制肝纤维化组织中α-SMA和ColⅠ的表达,因此可作为临床上预防和治疗肝纤维化的一种新型药物。 Objective To explore the effect of Lycium barbarum polysaccharide(LBP)on the expression of alpha-SMA and collagen I in hepatic fibrosis induced by common bile duct ligation in mice.Methods 30 healthy male mice weighing(0.32±0.28)kg were sorted into three groups by stochastic method:control group,model group and LBP intervention group,with an average of10 mice in each group.Model group:liver fibrosis was induced by ligation of common bile duct in mice.LBP intervention group:mice were given orally LBP 30μg/g on the 21 thday after ligation of common bile duct,once a day for 15 days.Control group:abdominal cavity was open,free common bile duct without ligation,and mice were given 0.9%sodium chloride solution at the same dose once a day for 15 days after closing the abdominal cavity.The liver tissue specimens of three groups were prepared.The pathological changes of liver tissue in three groups were analyzed by hematoxylin-eosin staining and Sirius red staining.The contents of alpha-SMA and type I collagen in liver tissue of three groups were determined by immunoblotting.Results In the model group,the structure of hepatic lobules was destroyed,liver tissue necrosis was found in different areas.The increase number of common bile ducts and fibrous tissue proliferation were found in the portal area,which indicated that common bile duct ligation could successfully induce hepatic fibrosis injury.The content of alpha-SMA and type I collagen in liver tissue(0.877±0.015,1.102±0.063)was raised as compared with the control group(0.286±0.005,0.765±0.017),and the differences were statistically significant(both P<0.05).In LBP intervention group,the necrosis of liver tissue,the number of common bile duct and the proliferation of fibrous tissue were reduced,and the contents of alpha-SMA and colla gen I in liver tissue(0.302±0.011,0.456±0.010)were decreased,and the difference was statistically significant compared with the model group(P<0.05).Conclusion LBP inhibits the expression of alpha-SMA and type I collagen in liver fibrosis tissue of mice,so it can be used as a new drug to prevent and treat liver fibrosis in clinic.
