摘要
植物源蛋白肽具有较好的抗氧化和降血压活性,研究采用蛋白酶Alcalase水解小米米糠分离蛋白(MBPI)获得小米米糠蛋白水解物(MBPH),经膜分离技术得到成4种不同分子质量大小的多肽组分。通过氧自由基吸附能力(ORAC)、血管紧张素转化酶(ACE)抑制活性、肾素(renin)抑制活性和体内降血压活性,评价了小米米糠蛋白水解物及其不同分子质量组分抗氧化及降血压活性,鉴定了<1 ku组分多肽的氨基酸序列。结果显示,低分子质量小米米糠蛋白水解物具有更好的氧自由基吸附能力和降血压活性,<1 ku组分的ORAC值为1455.41μmol TE/mg,ACE抑制率分为73.667%,肾素抑制率为74%,且在灌胃6 h后降低SHRs的收缩压约37.8 mm Hg;<1 ku组分中鉴定出7个小肽序列VALVR、VLER、VVRP、WVGK、FGPK、VLLF和LFGK。研究认为小米米糠蛋白水解物,尤其是<1 ku肽组分可能作为功能性成分用于降血压相关的功能食品和保健品的开发。
Plant-derived protein peptides are provided with good antioxiunt and antihypertensive activities.In this study,millet bran protein(MBPI)was digested with Alcalase to produce millet bran protein hydrolysate(MBPH),which was separated into different peptide fractions by membrane separation technology.Antihypertensive properties of MBPH and its different molecular weight peptide fractions were evaluated by analyzing their oxygen radical absorbance capacity(ORAC)、angiotensin converting enzyme-I(ACE)inhibitory activity、renin inhibitory activity and blood pressure in vivo activity,and the structure of the peptides were determined.The results indicated that the alkaline protease hydrolysate(<1 ku)of low molecular weight millet bran had better oxygen radical absorbability and antihypertensive activity.The<1 ku fraction had an ORAC value of 1455.41μmol TE/mg,ACE inhibition rate of 73.667%,renin inhibition rate of 74%and blood pressure of SHRs decreased to about 37.8 mm Hg after 6 hours of intragastric administration.Furthermore,the structure of<1 ku fraction was identified,and seven peptide sequences(VALVR、VLER、VVRP、WVGK、FGPK、VLLF and LFGK)were identified.In summary,membrane separation fractions of millet bran protein hydrolysate<1 ku may be used as functional components in the development of functional foods and health products related to lowering blood pressure.
