摘要
目的探讨超微川芎茶调散对偏头痛模型大鼠血浆β-EP、CGRP的影响。方法采用皮下注射硝酸甘油的方法构建偏头痛大鼠模型,将大鼠随机分为模型组、川芎茶调口服液组和超微川芎茶调散组,每组10只,另选取10只大鼠作为空白对照组(注射等体积生理盐水)。建模后0.5 h,川芎茶调口服液组(灌服川芎茶调口服液0.61 mL/kg),超微川芎茶调散组(灌服超微川芎茶调散0.12 g/kg),空白对照组和模型组(灌服等体积生理盐水),连续干预6 d。观察大鼠耳红出现时间、消退时间及挠头次数,比较各组大鼠血浆β-EP、CGRP含量。结果与模型组相比,川芎茶调口服液组和超微川芎茶调散组大鼠耳红消失时间缩短,且超微川芎茶调散组短于川芎茶调口服液组;T1~T7时间点川芎茶调口服液组和超微川芎茶调散组挠头次数少于模型组,超微川芎茶调散组少于川芎茶调口服液组;川芎茶调口服液组和超微川芎茶调散组血浆CGRP含量低于模型组,超微川芎茶调散组低于川芎茶调口服液组,超微川芎茶调散组β-EP含量高于模型组(均P<0.05)。结论川芎茶调散通过降低偏头痛发作时CGRP含量,升高β-EP含量,缓解神经源性炎症,发挥治疗偏头痛的作用,且超微川芎茶调散的效果优于川芎茶调口服液。
Objective To investigate the effect of superfine Chuanxiong Chatiao powder onβ-EP and CGRP in plasma of migraine model rats.Methods The model of migraine rats were established by subcutaneous injection of nitroglycerin.The rats were randomly divided into model group,Chuanxiong Chatiao oral liquid group and superfine Chuanxiong Chatiao powder group,with 10 rats in each group.Another 10 rats were selected as the blank control group(injected with equal volume of normal saline).After 0.5 h of modeling,Chuanxiong Chatiao oral liquid group(injected with 0.61 mL/kg of Chuanxiong Chatiao oral liquid),superfine Chuanxiong Chatiao powder group(injected with 0.12 g/kg of superfine Chuanxiong Chatiao powder),blank control group and model group(injected with equal volume of normal saline)were continuously intervened for 6 days.The occurrence time,resolution time and head scratching times of ear red in rats were observed,the expression ofβ-EP and CGRP in plasma of rats in each group were compared.Results Compared with the model group,the time of ear red disappeared in the Chuanxiong Chatiao oral liquid group and superfine Chuanxiong Chatiao powder group shortened,and the superfine Chuanxiong Chatiao powder group was shorter than the Chuanxiong Chatiao oral liquid group;the number of scratching heads in the Chuanxiong Chatiao oral liquid group and superfine Chuanxiong Chatiao powder group were less than the model group at T1~T7 time points,the superfine Chuanxiong Chatiao powder group was less than the Chuanxiong Chatiao oral liquid group;The plasma CGRP content of the Chuanxiong Chatiao oral liquid group and superfine Chuanxiong Chatiao powder group were lower than the model group,superfine Chuanxiong Chatiao powder group was lower than Chuanxiong Chatiao oral liquid group;the plasmaβ-EP of the superfine Chuanxiong Chatiao powder group was higher than the model group(all P<0.05).Conclusion Superfine Chuanxiong Chatiao powder can alleviate the neurogenic inflammation by reducing the content of CGRP and increasing the content ofβ-EP during migraine attack,so as to play a role in the treatment of migraine,and the effect of Superfine Chuanxiong Chatiao powder is better than that of Chuanxiong Chatiao oral liquid.
