摘要
肠道类器官是一种来源于肠道干细胞或多能干细胞(PSC)且具备三维结构的微器官。将肠道隐窝或单个干细胞植入基质胶中,在含有多种生长因子的培养皿中生成微型肠器官。该类器官包含各种肠道上皮细胞且能够有效地模拟肠组织。自2009年首次构建出肠道类器官,就迅速成为肠道疾病研究领域的热点。本文将围绕肠道类器官的构建及应用的最新进展进行述评。
Gut organoids are three-dimensional mini-guts that are derived from gut stem cells or pluripotent stem cells(PSC).Gut single crypts or stem cells,upon embedment into matrigel,are cultured under specific growth factors into self-organizing or⁃ganotypic structures.Organoids are comprised of all maj or types of gut epithelial cells and contain varying levels of physiologic similarity to native organ systems.Since first reported in 2009,gut organoids have soon become an important topic in the field of gut diseases.In this review article,we summarize the research advances in establishment and application of gut organoids.
