2种炮制方法对女贞子多酚含量及脂肪酸组成的影响

Effects of Two Types of Processing Methods on Polyphenols and Fatty Acid Composition of Ligustrum Lucidum

摘要 目的:探究不同炮制方法对中药女贞子中多酚含量及脂肪酸组成的影响。方法:采用酒制和醋制两种方法对女贞子进行炮制,炮制时间为1~8h;采用超声法提取女贞子多酚,运用Folin酚法测定多酚含量;采用索氏法提取脂肪酸,经甲酯化后运用GS-MS分析脂肪酸组成。结果:酒制后多酚含量为611.71~572.77mg/L,在酒制4h时多酚的含量达到最大;醋制后多酚含量为599.93~568.69mg/L,经过醋制后多酚含量低于生品。女贞子生品与炮制品中油脂均主要由不饱和脂肪酸组成。生品女贞子中不饱和脂肪酸占总脂肪酸的84.31%,其中油酸占49.6%,油酸/亚油酸(O/L)比值为1.56。酒制女贞子中不饱和脂肪酸占总脂肪酸的84.27%~84.89%,其中油酸占50.43%~53.42%,O/L比值为1.60~1.72。醋制女贞子中不饱和脂肪酸占总脂肪酸的84.27%~85.43%,其中油酸占50.33%~52.59%,O/L比值为1.58~1.71。结论:女贞子经过炮制后,其多酚含量与脂肪酸组成均有不同程度的变化,其中酒制4h是较为适宜的炮制方法。 Objective:To explore the effects of different processing methods on the content of total polyphenols and fatty acid composition in Ligustrum Lucidum.Methods:The Ligustrum Lucidum was processed by alcohol and vinegar,and the time of processing was from 1 h to 8 h.The polyphenol was obtained by ultrasonic extraction,and determined by Lowry method.Fatty acid was extracted by Soxhlet method,and its composition was analyzed by GS-MS after methyl esterification.Results:The polyphenol content of Ligustrum Lucidum after processed by using alcohol and vinegar are 611.71~572.77 mg/L and 599.93~568.69 mg/L,respectively.The polyphenol content reached the maximum at 4 h by using alcohol,and was lower than unprocessed Ligustrum Lucidum.The unsaturated fatty acids in the unprocessed Ligustrum Lucidum accounted for 84.31%of the total fatty acids,of which oleic acid accounted for 49.6%,and its Oleic acid/linoleic acid(O/L)ratio is 1.56.The unsaturated fatty acids in the processes by alcohol accounted for 84.27%~84.89%of the total fatty acids,of which oleic acid accounted for 50.43%~53.42%,and its O/L ratio is 1.60~1.72.The unsaturated fatty acids in the processes by vinegar accounted for 84.27%~85.43%of the total fatty acids,of which oleic acid accounted for 50.33%~52.59%,and its O/L ratio is 1.58~1.71.Conclusion:After processing,the polyphenol content and fatty acid compositions of Ligustrum Lucidum have changed to varying degrees,and the 4 h of processes by alcohol is a suitable method.
