电针“解郁方”对抑郁症大鼠模型血清中吲哚胺2,3双加氧酶、5-羟色胺、白介素10含量的影响 被引量:4

摘要 目的:观察电针“解郁方”(百会、神门、太冲)对抑郁症大鼠模型血清中吲哚胺2,3双加氧酶(IDO)、5-羟色胺(5-HT)、白介素10(IL-10)含量的影响。方法:按照随机数字表将40只清洁级SD雄性大鼠随机分为空白组、模型组、假针组、电针组4个组,每组10只,所有大鼠均是经过旷场实验筛选出来的,采用束缚3 h,夹尾巴3 min,热烘5 min(45~60℃),禁食、禁水各24 h,昼夜颠倒24 h,4℃下冰水游泳5 min,再配合潮湿垫料造慢性不可预测应激抑郁症大鼠模型,每天随机安排一种造模方法造模,每种方法使用4次,共造模28 d。空白组正常喂养,模型组在假针组、电针组治疗时只抓取固定。电针组和假针组于造模开始即治疗,28 d后麻醉腹主静脉采血,ELISA双抗夹心法检测血清IDO、5-HT、IL-10含量。结果:与空白组比较,模型组血清中IDO含量显著增加(P<0.01),5-HT、IL-10含量明显下降(P<0.01);与模型组比较,假针组和电针组血清IDO含量均降低(P<0.01),5-HT、IL-10均上升(P<0.01);与假针组比较,电针组血清IDO含量下降(P<0.05),5-HT含量显著增加(P<0.01),IL-10含量上升(P<0.05),差异有统计学意义。结论:电针调节血清中的IDO、5-HT、IL-10含量可能是其治疗抑郁症的机制之一。 Objective:To explore the mechanism of electroacupuncture on“Jieyufang”(Baihui,Shenmen,and Taichong)on rats with depression.Methods:According to the random number table,40 clean SD male rats were randomly divided into four groups:blank group,model group,sham needle group,and electroacupuncture group.Each group had 10 rats.All rats were selected through open field experiments.Modeling method of chronic unpredictable stress depression rat model included restraining for 3 h,clipping tail for 3 min,hot baking at 45~60℃for 5 min,fasting and water prohibition for 24 h,day and night upside down 24 h,swimming in 4℃ice water for 5 min and using damp padding,and a modeling method was randomly arranged every day.Each method was used 4 times in 28 days.The blank group was fed normally,and the model group was grasped and fixed during the treatment of the sham needle group and the electroacupuncture group.The electroacupuncture group and the sham needle group started treatment at the same time of molding.After 28 days,blood was collected from the abdominal main vein after anaesthesia,and then contents of IDO,5-HT and IL-10 in serum were measured by ELISA double antibody sandwich method.Results:Compared with the blank group,the IDO content in model group was increased significantly(P<0.01),and the levels of 5-HT and IL-10 were decreased significantly(P<0.01).Compared with the model group,IDO content was decreased(P<0.01)while contents of 5-HT and IL-10 were significantly increased(P<0.01)in the sham needle group and the electroacupuncture group(P<0.01).Compared with sham needle group,IDO content was decreased(P<0.05)and contents of 5-HT(P<0.01)and IL-10 were increased(P<0.05)in electroacupuncture group,the difference was statistically significant.Conclusion:Electroacupuncture plays an antidepressant role by regulating contents of IDO,5-HT,and IL-10 in serum of chronic unpredictable stress depression rat model.
