摘要
本研究利用Illumina HiseqTM4000测序平台对柚木(Tectona grandis L.F.)边材组织进行转录组测序,获得39.65 Gb的数据。拼接组装共得到90843个Unigene,平均长度、N50以及GC含量分别为1415 bp,2208 bp和41.28%。将获得的Unigene与七大功能数据库进行比对,分别有64416(NR:70.91%)、69281(NT:76.26%)、28777(COG:31.68%)、18630(GO:20.51%)、49594(KEGG:54.59%)、44707(Swissprot:49.21%)以及50938(Interpro:56.07%)个Unigene获得功能注释。经过GO数据库的比对分析,18630个Unigene被注释到生物过程、细胞组分和分子功能3大类别55个亚类。与COG数据库进行比对分析,28777个注释Unigene按功能被划分为25类。基于KEGG数据库,44595个Unigene序列注释到6大类,21个亚类代谢通路中。根据注释结果预测出2772个编码转录因子的Unigene,检测出26773个SSR位点,以及39856个SNP位点。本研究为柚木分子育种工作的开展提供数据和参考。
In t his study,we obtained about 39.65 Gb data in total from sapwood xylem transcriptome of teak(Tectona grandis L.F.)by Illumina HiseqTM4000 sequencing.After assembling,90843 Unigene were obtained,the average length,N50,and GC content of Unigenes were 1415 bp,2208 bp,and 41.28%,respectively.Then,the Unigenes were annotated by 7 functional databases,and 64416(NR:70.91%),69281(NT:76.26%),28777(COG:31.68%),18630(GO:20.51%),49594(KEGG:54.59%),44707(Swissprot:49.21%)and 50938(Interpro:56.07%)Unigenes were annotated finally.GO analysis showed that,18630 Unigenes divided into 55 sub-categories of three functional categories(biological process,cellular components and molecular function).Compared with the COG database,28777 Unigenes were assigned to 25 categories according to the function.The 44595 Unigenes were grouped into 6 major classes of 21 branches based on the KEGG database.The 2772 Unigenes encoding transcription factors(TF)were predicted with functional annotation results.Meanwhile,26773 SSR markers and 39856 SNP polymorphic loci were detected.This transcriptome sequencing analysis could lay a foundation for molecular breeding of Tectona grandis.
