摘要
目的通过现代生物信息挖掘技术,探究淫羊藿对高脂血症的作用靶标及参与的生物学过程,并通过动物实验观察淫羊藿降血脂效果。方法借助TCMIP 2.0 整合药理学平台,检索淫羊藿的所有成分及靶点,并获得高脂血症的疾病所有靶标,构建蛋白-蛋白相互作用(PPI)网络,富集分析药物干预疾病关键靶标,取前二十分析淫羊藿在治疗高脂血症中的生物过程及作用途径,通过高脂喂养大鼠,给予淫羊藿,检测大鼠血脂,进行病理切片的HE染色。结果对于淫羊藿治疗高脂血症共获得23个关键核心靶标,其中药物与疾病共有靶标2个,分别为 PPARγ和MTTP,靶标富集的生物学过程主要与蛋白质的活化,能量的代谢等途径相关,淫羊藿对高脂饮食大鼠血脂具有降低作用,对肝脏具有保护作用。结论淫羊藿在治疗高脂血症方面具有一定的作用,可对脂质代谢产生调控。
Objective To explore the targets and biological processes of Epimedium brevicornu Maxim in the treatment of hyperlipidemia based on modern biological information mining technology.The effect of Epimedium brevicornu Maxim on lowering blood lipid was observed by animal experiments.Methods Using the TCMIP 2.0 integrated pharmacological platform to retrieve all components and targets of Epimedium brevicornu Maxim and to obtain all targets for Hyperlipidemia diseases to construct a protein-protein interaction(PPI)network,and to enrich and analyze key targets for drug intervention in diseases,and then to take the first twenty to analysis of the biological process and effects of Epimedium brevicornu Maxim in the treatment of Hyperlipidemia.The rats were fed with high fat and given Epimedium brevicornu Maxim,their blood lipids were detected,and the pathological sections were stained with HE.Results A total of 23 key core targets of Epimedium brevicornu Maxim for the treatment of Hyperlipidemia were obtained,of which there were 2 targets for drugs and diseases,respectively PPARγand MTTP.Epimedium brevicornu Maxim can reduce the blood lipid and protect the liver in high fat diet rats.Conclusion Epimedium brevicornu Maxim has a certain effect in the treatment of Hyperlipidemia and can regulate lipid metabolism.
