摘要
目的研究食管癌患者鳞状细胞癌抗原(SCC)、癌胚抗原(CEA)、细胞角蛋白19片段(CYFRA21-1)的表达水平,并分析其临床意义。方法选取2016年5月至2018年5月安康市中心医院收治的食管癌患者100例,选取同期健康成年人100例为对照。比较两组研究对象血清SCC、CEA、CYFRA21-1水平的差异,比较不同临床病理特征食管癌患者血清SCC、CEA、CYFRA21-1水平的不同,采用线性回归分析影响食管癌患者SCC、CEA、CYFRA21-1水平的因素。结果食管癌组的血清SCC、CEA、CYFRA21-1水平分别为(2.98±0.87)μg/ml、(1.95±0.34)μg/L、(2.86±0.47)ng/ml,分别高于对照组的(1.54±0.42)μg/ml、(0.86±0.25)μg/L、(1.43±0.29)ng/ml,差异有统计学意义(t=14.906、25.828、25.893,P<0.001);分化程度为中低分化、有淋巴结转移、TNM分期为Ⅲ+Ⅳ期食管癌患者的血清SCC、CEA、CYFRA21-1水平较高,不同年龄、性别的食管癌患者血清SCC、CEA、CYFRA21-1水平无明显差异;线性回归分析显示肿瘤分化程度和TNM分期是影响食管癌患者SCC、CEA、CYFRA21-1水平的因素。结论食管癌患者血清SCC、CEA、CYFRA21-1水平较高,且受肿瘤分化程度和TNM分期的影响。
Objective To explore the role mechanism of lentinan(LNT)on sensitization of paclitaxel(PTX)chemotherapy of human breast cancer cell line MCF-7.Methods Human breast cancer MCF-7 cell strains were randomly divided into control group,LNT group,PTX group and LNT+PTX group.PTX group was given 100μl of PTX(4 nmol/L),and LNT group was given 100μl LNT(0.1μmol/L),and LNT+PTX group was given 50μl of PTX and LNT,and they were incubated for 24 h with cells.Control group was only added RPMI 1640 medium and the same volume of DMSO.After 24 h of culture,the cell inhibition rate was detected by MTT assay,and the apoptosis was detected by TUNEL method,and the expression levels of apoptosis-related proteins Caspase-3,Bcl-2 and Bax were detected by Western blot.Real-time quantitative(RT)-PCR was used to detect the expression levels of resistance-related genes MDR1,MRP1 and BCRP.Results The apoptosis rate of MCF-7 cells in PTX group was significantly higher than control group(P<0.05).The inhibition rate and apoptosis rate in LNT+PTX group was significantly higher than PTX group(P<0.05),and the IC50 was significantly lower than PTX group(P<0.05).The protein expression levels of Caspase-3 and Bax in PTX group and LNT+PTX group were significantly higher than control group(P<0.05),and the expression level of Bcl-2 was significantly lower than control group(P<0.05),and there were no significant differences in the expression levels of resistant genes MDR1,MRP1 and BCRP between PTX group and control group(P>0.05),and the expression levels of resistant genes MDR1,MRP1 and BCRP in LNT group and LNT+PTX group were significantly lower than control group(P>0.05).The protein expression levels of Caspase-3 and Bax in LNT+PTX group were significantly higher than PTX group(P<0.05),and the expression levels of Bcl-2 and resistance genes MDR1,MRP1 and BCRP were significantly lower than PTX group(P<0.05).Conclusions LNT inhibits the expression of resistance genes MDR1,MRP1 and BCRP,enhance the sensitivity of PTX in MCF-7 cells,and promote the cell inhibition and apoptosis of PTX.
