黑线仓鼠GAL克隆及生物信息学分析

摘要 以黑线仓鼠(Cricetulus barabensis)为对象,采用RT-PCR技术克隆黑线仓鼠GAL cDNA序列482 bp,采用生物信息分析软件对GAL进行基因序列分析表明,结果显示GAL包括完整CDS区375 bp和部分5'和3'非编码区,共编码124个氨基酸,GAL蛋白质分子量为13199.05 Da,其不稳定指数为44.67,高于40,表明GAL蛋白不稳定.GAL含有信号肽,为分泌蛋白.同源性比对及系统进化树分析,数据显示黑线仓鼠(Cricetulus barabensis)与中国仓鼠(Cricetulus griseus)亲缘关系相对较近,而与啮齿类等亲缘关系次之,与灵长类及其他鸟类物种亲缘关系较远,这与传统的物种分类较为一致,进而验证克隆所得到的GAL cDNA序列准确,体现了GAL进化的保守性,为探究GAL在动物繁殖调控中的作用提供了一定的理论基础. This study used the striped hamsters Cricetulus barabensis as the research materials,GAL cDNA sequence 482 bp of the striped hamster was cloned by RT-PCR.the biological information analysis software of GAL gene sequence analysis,results show that the GAL including full CDS area 375 bp and part 5′and 3′noncoding region,encoding 124 amino acids,the molecular weight of GAL protein was 13199.05 Da,and its instability index was 44.67,higher than 40.GRAVY value is-0.078,indicating that GAL protein is unstable.GAL is a secretory protein that contains signal peptides.The homology comparison and phylogenetic tree analysis showed that the striped hamster and the Chinese hamster were closely related.Secondly,the striped hamster is closely related to rodents and is far related to primates and other bird species,which is consistent with previous species classification.Furthermore,it was verified that the sequences obtained by cloning were accurate and it refle the conservatism of CAL evolution.This study provides the theoretical basis for exploring the role of GAL in the regulation of animal reproduction and is of great significance.
