摘要
为研究兰科天麻属植物天麻(Gastrodia elata)块根中内生真菌(Penicillium skriabinii)的次生代谢产物,菌株采用固体发酵,发酵物用甲醇超声提取,综合运用硅胶柱色谱、LH-20羟丙基葡聚糖凝胶柱色谱(sephadex LH-20)及反相硅胶柱色谱等方法进行分离和纯化,得到12个单体化合物。根据化合物的理化性质及波谱数据鉴定其结构,分别为:preaustinoid A1(1),preaustinoid A2(2),1,2-dihydro-acetoxydehydroaustin(3),dehydroausin(4)、austin(5),verruculogen(6),(5aS,12S,14aS)-5a,6-dihydroxy-12-(2-hydroxy-2-methylpropyl)-9-methoxy-1,2,3,5a,6,14a-hexahydropyrrolo[1″,2″:4′,5′]pyrazino[1′,2′:1,6]pyrido[3,4-b]indole-5,14(11H,12H)-dione(7),cyclotryprostatin A(8),cyclo-L-tryptophyl-L-proline(9),6-methoxy-5(6)-dihydropenicillic acid(10),青霉酸(11)和β-谷甾醇(12)。
To study the secondary metabolites of Penicillium skriabinii,an endophytic fungus derived from the roots of Gastrodia elata.The strain was cultured in solid fermentation,and soaked in methanol via ultrasonic extraction.Twelve secondary metabolites were isolated and purified from its culture by combination of silica gel column chromatography,sephadex LH-20 gel column chromatography,and reversed-phase silica gel chromatography.The structures were elucidated by the physicochemical properties and spectroscopic data to be preaustinoid A1(1),preaustinoid A2(2),1,2-dihydro-acetoxydehydroaustin(3),dehydroausin(4),austin(5),verruculogen(6),(5aS,12S,14aS)-5a,6-dihydroxy-12-(2-hydroxy-2-methylpropyl)-9-methoxy-1,2,3,5a,6,14a-hexahydropyrrolo[1″,2″:4′,5′]pyrazino[1′,2′:1,6]pyrido[3,4-b]indole-5,14(11H,12H)-dione(7),cyclotryprostatin A(8),cyclo-L-tryptophyl-L-proline(9),6-methoxy-5(6)-dihydropenicillic acid(10),penicillic acid(11),andβ-sitosterol(12).
