摘要
为研究褪黑素对干旱胁迫下大麦(Hordeum vulgare)生理及蜡质基因表达的影响,以野生型大麦品种‘浙农大3号’、蜡粉缺失突变体p1为试验材料,采用溶液培养法,在两叶一心期比较干旱胁迫下不同浓度(10、50和100μmol·L-1)褪黑素处理的大麦幼苗叶片的生理指标差异及蜡质基因的表达情况。结果表明,干旱胁迫下,随着褪黑素浓度依次升高,超氧化物歧化酶和过氧化物酶活性以及脯氨酸含量先上升后下降,丙二醛含量则先下降后上升。选取的蜡质相关基因中,在干旱胁迫下,50μmol·L-1 MT处理的MYB94、CER6、CER10和TA6-SFT基因上调。本研究揭示适宜浓度的褪黑素通过生理和分子水平提高大麦幼苗的抗旱能力,缓解干旱胁迫带来的伤害。
In order to study the effect of melatonin on physiology and wax gene expression of barley(Hordeum vulgare)under drought stress,the wild-type‘ZJU3’and wax-deficient mutant p1 were used as materials.The differences of physiological indexes and the expression of waxy genes of barley seedlings were compared treated with different concentrations(10,50 and 100μmol·L-1)of melatonin under drought stress.The results showed that under drought stress,with the increase of melatonin concentration,the activities of superoxide dismutase and peroxidase and proline content first increased and then decreased,while the malondialdehyde content first decreased and then increased.Among the wax related genes selected,MYB94,CER6,CER10 and TA6-SFT genes were up-regulated by adding 50μmol·L-1 MT under drought stress.This study revealed that the appropriate concentration of melatonin could improve the drought-resistant ability of barley seedlings at physiological and molecular levels,and alleviate the damage caused by drought stress.
