摘要
为研究亚甲基蓝(MB)对猪卵母细胞体外发育的影响,选取500个猪卵母细胞随机分为5组,各组添加亚甲基蓝浓度为0、10、50、100和200 nmol/mL,培养成熟44 h^46 h后,观察不同浓度的亚甲基蓝对卵母细胞成熟率影响;孤雌激活2 d^7 d后,观察胚胎卵裂率和囊胚率以及活性氧(ROS)、谷胱甘肽(GSH)、母系表达基因含量的变化。结果表明,亚甲基蓝能显著提高猪卵母细胞孤雌激活后胚胎卵裂率和囊胚率,提高孤雌胚中GSH含量,降低ROS含量,增加孤雌激活胚中母系基因细胞周期素B1表达量。结果提示,一定剂量的亚甲基蓝可有效改善猪卵母细胞在体外的发育能力。
To study the effect of methylene blue(MB)on the development of porcine oocytes in vitro,500 porcine oocytes were selected and randomly divided into five groups.These five groups of oocytes were respectively treated with different concentrations of MB with a gradient of 0,10,50,100 and 200 nmol/mL.After 44-46 hours of oocytes culture and maturation,the effect of different MB concentration on maturation rate of oocytes were recorded.Following with parthenogenetic activation for 2-7 days,the effects of embryonic cleavage rate,blastocyst rate,reactive oxygen species(ROS)content,glutathione(GSH)content and maternal gene expression level were respectively tested and recorded.The result indicated that methylene blue can significantly increase cleavage rate and blastocyst rate in parthenogenetic activated oocytes.Additionally,MB treatment can elevate GSH content and decrease ROS content in parthenogenetic embryos.It can also help to up-regulate B1 gene expression level,a maternal gene,in parthenogenetic activated oocytes.In summary,results of this study showed that a certain dose of methylene blue can effectively improve in-vitro developmental competence of porcine oocytes.
