摘要
目的:探讨柴胡三参胶囊对缺血性心律失常大鼠心肌细胞兰尼碱受体(ryanodine receptors,RyR) mRNA表达的影响,为柴胡三参胶囊的临床应用提供实验依据。方法:60只Wistar大鼠随机分为假手术组、模型组、空白组、柴胡三参胶囊组、参松养心胶囊组、维拉帕米组,每组10只。相应干预10 d后,空白组不作任何处理,假手术组只开胸不结扎,其余各组采取大鼠左冠状动脉前降支高位结扎法制作心肌缺血性心律失常模型,再用PCR法检测RyR2 mRNA在各组大鼠心肌中的表达水平情况。结果:柴胡三参胶囊组大鼠心律失常评分低于模型组(P<0.05);柴胡三参胶囊组大鼠死亡率低于模型组;柴胡三参胶囊组大鼠RyR2 mRNA CT值的相对表达量低于模型组(P<0.05)。结论:柴胡三参胶囊可通过抑制RyR2 mRNA的过度表达,减少缺血性心律失常的发生,从而降低大鼠的死亡率。
Objective:To explore the effect of Chaihu Sanshen capsule on the expression of ryanodine receptors(RyR)2 in myocardial cells of ischemic arrhythmia rats,and to provide experimental basis for the clinical application of Chaihu Sanshen capsule.Methods:A total of 60 Wistar rats were randomly divided into sham operation group,model group,blank group,Chaihu Sanshen capsule group,Shensong Yangxin capsule group and verapamil group,10 rats in each group.After corresponding intervention for 10 days,the blank group was left without any treatment,the sham operation group only had thoracotomy without ligation,and the other groupsused the high-level ligation of the left anterior descending coronary artery to make a myocardial ischemic arrhythmia model.The expression level of RyR2 mRNA in myocardium of rats in each group was detected by PCR.Results:The arrhythmia score of the rats in the Chaihu Sanshen capsule group was lower than that in the model group(P<0.05).The mortality rate of the rats in the Chaihu Sanshen capsule group was lower than that in the model group.The relative expression of CT value of RyR2 mRNA in Chaihu Sanshen capsule group was lower than that in model group(P<0.05).Conclusion:Chaihu Sanshen capsule can reduce the occurrence of ischemic arrhythmia by inhibiting the overexpression of RyR2 mRNA,thereby reducing the mortality of rats.
