摘要
目的探讨miR-93在胃癌患者血清中的表达变化及与临床病理特征及预后的关系。方法选取2015年10月至2016年5月接受手术治疗的胃癌患者88例为胃癌组,选择同期体检的60例健康人为对照组。采用实时荧光定量PCR(qRT-PCR)检测研究对象血清中miR-93表达水平,分析血清miR-93表达与胃癌患者临床病理特征及预后的关系,Cox风险回归分析影响胃癌患者预后的因素,采用ROC曲线分析血清miR-93水平对胃癌发生的诊断价值。结果胃癌组血清中miR-93相对表达水平明显高于对照组(P<0.05);血清中miR-93表达水平与肿瘤直径、病理分化程度、TNM分期、淋巴结转移密切相关(P<0.05);ROC曲线分析结果显示血清miR-93诊断胃癌发生的曲线下面积AUC为0.975(95%CI为0.960~0.991,P<0.05)。生存分析显示血清miR-93高表达组患者预后较差(P<0.05);Cox风险回归分析结果显示淋巴结转移及血清miR-93表达水平是影响胃癌患者预后的独立因素(P<0.05)。结论血清中的miR-93表达水平升高与胃癌的发生、发展有关,血清miR-93可能成为胃癌诊断及预后判断的生物学标志物。
Objective To investigate the changes of serum levels of miR-93 in patients with gastric cancer(GC)and its correlation with clinicopathological features and prognosis of patients.Methods A total of 88 patients with GC who were treated by surgery in our hospital from October 2015 to May 2016 were enrolled as GC group,and theother 60 healthy subjects who underwent physical examination in our hospital at the same period were enrolled as control group.The qRT-PCR was used to detect the expression levels of miR-93 in the serum of the subjects.The correlation between the expression levels of miR-93 in serum and the clinicopathological characteristics as well as the prognosis of patients with GC was analyzed.Moreover Cox risk regression analysis was used to analyze the prognostic factors of patients with GC,and ROC curve was used to analyze the diagnostic value of miR-93 levels detection in the occurrence of GC.Results The relative expression levels of miR-93 in GC group were significantly higher than those in control group(P<0.05),and the expression levels of serum miR-93 were closely correlated with tumor diameter,pathological differentiation degree,TNM stage and lymph node metastasis(P<0.05).ROC curve analysis showed that the area under the curve of serum microRNA-93 in diagnosis of gastric cancer was 0.975(95%CI was 0.964~0.991,P<0.05).Survival analysis showed that the prognosis of patients with high expressions of serum miR-93 was poor(P<0.05).Cox risk regression analysis showed that lymph node metastasis and serum miR-93 expression were the independent factors affecting the prognosis of patients with GC(P<0.05).Conclusion The increase of expression levels of serum miR-93 is related to the occurrence and development of GC,and the serum miR-93 may be regarded as a biomarker for the diagnosis and prognosis evaluation in patients with GC.
