五子衍宗丸对帕金森病小鼠的神经保护及抗炎作用 被引量:14

Neuroprotective and anti-inflammatory effects of Wuzi Yanzong Pills on Parkinson’s disease mice

摘要 目的:探析五子衍宗丸对帕金森病(PD)小鼠的神经保护及抗炎作用。方法:24只C57BL/6雄性小鼠随机分为模型组和五子衍宗丸组,每组12只。腹腔注射MPTP建立小鼠PD模型,同时模型组灌胃0.9%氯化钠溶液,五子衍宗丸组灌胃五子衍宗丸溶液,持续2周。采用旷场和步态实验观察小鼠行为学表现;免疫组织化学染色观察中脑黑质TH阳性神经元;ELISA检测脑组织中细胞因子IL-6、TNF-α、IL-1β、IL-10水平。结果:与模型组比较,五子衍宗丸组小鼠移动总距离及平均速度均显著增加(P<0.01),休息时间和脚步角度显著减少(P<0.05),自主活动量增多;形态学显示中脑黑质TH阳性细胞数显著增多(P<0.05);炎性因子IL-1β、IL-6、TNF-α水平显著降低(P<0.05,P<0.01),抗炎因子IL-10水平显著升高(P<0.01)。结论:五子衍宗丸可明显改善PD小鼠的行为学,保护黑质多巴胺能神经元,其作用机制可能与抑制脑内炎症反应有关。 Objective:To explore the neuroprotective and anti-inflammatory effect of Wuzi Yanzong Pills(WYP)in MPTP-induced Parkinson’s disease(PD)mice.Methods:Twenty-four C57 BL/6 male mice were randomly divided into the model group and WYP group,12 in each group.Mice PD model was established by intraperitoneal injection of MPTP for one week.At the same time,the model group was given 0.9%sodium chloride solution,the WYP group was given WYP solution,lasting two weeks.The animal behavior was evaluated by open field and gait.TH^+ neuron in the substantia nigra of the midbrain was measured by immunohistochemistry.The levels of cytokine IL-6,IL-10,IL-1β,and TNF-α in the extract of the brain were detected by ELISA.Results:Compared with the model group,the total distance and average speed of movement in the WYP group increased significantly(P<0.01),while the rest time and step angle decreased obviously(P<0.05),and the amount of autonomous activity increased.In substantia nigra of bran,TH^+ neuron increased significantly after WYP intervention(P<0.05).At the same time,the levels of inflammatory factors IL-1β,IL-6,and TNF-α decreased significantly(P<0.05,P<0.01),while anti-inflammatory factor IL-10 increased significantly(P<0.01).Conclusion:WYP can significantly improve the behavior of PD mice and protect dopaminergic neurons,which may be related to the anti-inflammatory effect of WYP.
