沉默ENO1对人宫颈癌HeLa细胞增殖、迁移、侵袭能力的影响 被引量:1

摘要 目的探讨α-烯醇化酶(ENO1)在宫颈癌组织中的表达及其对宫颈癌HeLa细胞增殖、迁移和侵袭的影响。方法运用免疫化学染色和实时荧光定量PCR(qRT-PCR)检测48例宫颈癌患者的癌组织和癌旁组织中ENO1的表达情况。运用RNA干扰技术对人宫颈癌HeLa细胞中的ENO1进行干扰,将细胞分为对照组(PBS组)、空载体组(GV102组)和干扰组(shENO1组),采用RT-PCR和Western blot检测干扰效率。通过MTT实验、细胞划痕实验和Transwell实验检测ENO1对细胞增殖、迁移和侵袭的影响。结果ENO1在宫颈癌组织中高表达(P<0.001),与年龄、FIGO分期、淋巴转移、病理类型及分级无明显相关性(P>0.05)。shENO1组ENO1 mRNA和蛋白表达水平较PBS组和GV102组显著降低(P<0.05)。MTT实验、细胞划痕实验和Transwell实验结果显示,shENO1组细胞增殖、迁移和侵袭能力较PBS组和GV102组显著降低(P<0.05)。结论ENO1在宫颈癌组织中呈高表达,体外实验发现沉默ENO1能抑制宫颈癌HeLa细胞的增殖、迁移和侵袭能力。 Objective To investigate the expression ofα-enolase(ENO1)in cervical cancer and its effect on the proliferation,the migration and the invasion of cervical cancer HeLa cells.Methods The expression of ENO1 was detected in cancer and adjacent tissues from 48 patients with cervical cancer by immunochemical staining and real-time fluorescent quantitative PCR(qRT-PCR).HeLa cells were divided into three groups:control group(PBS group),empty vector group(GV102 group)and interference group(shENO1 group).The interference efficacy was verified by RT-PCR and Western blot.Meanwhile,MTT assay,cell scratch assay and Transwell assay were used to detect the effects of ENO1 on the cell proliferation,the migration and the invasion.Results ENO1 expression was higher in cervical cancer tissues than in adjacent tissues(P<0.001).But the high expression of ENO1 was not related to age,FIGO stage,lymph node metastasis,pathological type and grade(P>0.05).Compared with PBS group and GV102 group,the expression levels of ENO1 mRNA and protein were decreased significantly in shRNA group(P<0.05).The capacities of cell proliferation,migration and invasion in shENO1 group were significantly lower than those in PBS group and GV102 group(P<0.05).Conclusion ENO1 is expressed highly in cervical cancer tissues.The ENO1 silencing could inhibit the proliferation,the migration and the invasion of cervical cancer HeLa cells.
