Extraction Optimization of Total Flavonoids in Hippophae Rhamnoides L.by Central Composite Design and Response Surface Method

摘要 目的:优化沙棘总黄酮提取工艺,获取沙棘总黄酮最佳提取工艺条件及技术参数。方法:采用Box-Benhnken中心组合设计,以提取物中总黄酮含量和总黄酮提取率的总权重评分值为评价指标,对提取液料比、乙醇浓度、提取时间3个影响因素进行考察,以Design-Expert 8.05软件对试验数据作统计处理。结果:优化得到最佳工艺条件:9倍量70%乙醇,回流提取2次,每次2 h。结论:优选得到的工艺稳定可行,可作为沙棘提取提供参考依据。 Objective:To optimize the extraction of total flavonoids in Hippophae rhamnoides L.and obtain the optimal extraction conditions and technical parameters of total flavonoids in Hippophae rhamnoides L.Methods:With the content and the extraction rate of total flavonoids in Hippophae rhamnoides L.as the indices,the effects of three factors including the solvent-solid ratio,ethanol concentration and extraction time were explored by Box-Behnken central composite.Design-Expert 8.05 software was used to statistically process the experimental data.Results:The optimal extraction conditions of Hippophae rhamnoides L.were as follows:the ethanol concentration was 90%,the extracting time was two hours and the solvent-solid ratio was 9.Conclusion:The optimized extraction process of total flavonoids is stable and feasible,and the result can provide theoretical guidance for the extraction of total flavonoids in Hippophae rhamnoides L.
