叶生小皮伞不同发酵产物中麦角甾醇和麦角甾酮的含量分析 被引量:1

摘要 为了分析叶生小皮伞菌株ME-1通过不同发酵方式得到的发酵产物中麦角甾醇和麦角甾酮2种活性成分的含量,采用HPLC双波长法测定液体发酵菌丝体和固体发酵产物中的麦角甾醇和麦角甾酮含量。结果表明:麦角甾醇、麦角甾酮分别在0.1~10.0和0.0123~0.1230 g,线性良好(R^2=0.9998、R^2=0.9996),精密度、稳定性、重复性的RSD均小于2.0%,加样回收率在99.30%~103.93%范围内;含量分析表明,液体发酵菌丝体中麦角甾醇含量为125.0μg/g,麦角甾酮含量为1.2μg/g;固体发酵物中麦角甾醇的含量为37.7μg/g,麦角甾酮的含量为4.8μg/g。由此看出,利用液体发酵方式可在其菌丝体中获得更多的麦角甾醇,而固体发酵产物中的麦角甾铜含量较高。本研究建立了稳定、高效、重现性好的叶生小皮伞活性成分的HPLC双波长分析法,明确了不同发酵方式下不同发酵产物中麦角甾醇和麦角甾酮的差异,为叶生小皮伞的综合评价和质量控制提供了理论依据。 In order to analysis the content of ergosterol and ergosterone in the fermentation products obtained from Marasmius epiphyllus strain ME-1 by different fermentation methods,the content of ergosterol and ergosterone in liquid fermentation mycelium and solid fermentation product was determined by HPLC dual-wavelength method.The results showed that the ergosterol and ergotsterone were well separated with good linear correlations(R^2=0.9998,R^2=0.9996)in the range of 0.1-10.0 and 0.0123-0.1230 g.RSDs of precision,stability and repeatability tests were all less than 2.0%,the average recoveries were 99.30%-103.93%.The analysis results showed that the content of ergosterol and ergotsterone of mycelium in liquid fermentation was 125.0 and 1.2μg/g,respectively,the content of ergosterol and ergotsterone of solid fermentation products was 37.7 and 4.8μg/g.The study showed that more ergosterol could be obtained from the liquid fermentation,while more ergotsterone from the solid fermentation.The HPLC with dual wavelength analysis method with sable,efficient and good reproducibility was successfully established,the difference of the content of ergosterol and ergotsterone in different fermentation products under different fermentation methods was clarified.It provides the theoretical basis for the comprehensive evaluation and quality control of Marasmius epiphyllus.
