摘要
目的:探讨内热针治疗对第三腰椎横突综合征大鼠损伤组织炎症反应及Akt/NF-κB信号通路的影响。方法:将90只SD大鼠按随机数字表法分为正常组、模型组、药物组、内热针组、普通针刺组和温针灸组6组各15只,采用埋置明胶海绵的方法制作第三腰椎横突综合征模型,2周后进行为期4周的治疗。治疗后,颈静脉取血检测血清中TNF-α、IL-1β、IL-6和IL-8的含量,HE染色观察第三腰椎横突肌肉组织病理学变化以及Western blot检测损伤组织中p-Akt(S473)和NF-κB(p65)蛋白表达情况。结果:与正常组比较,模型组血清中TNF-α、IL-1β、IL-6和IL-8的含量显著升高,损伤组织中炎症反应严重,且p-Akt(S473)和NF-κB(p65)蛋白上调显著。与模型组比较,各治疗组血清TNF-α、IL-1β、IL-6和IL-8的含量降低,组织炎症反应减轻,p-Akt(S473)和NF-κB(p65)蛋白水平下降,且内热针组效果最明显。结论:内热针治疗可通过抑制Akt/NF-κB信号通路的激活,降低炎性细胞因子表达,改善第三腰椎横突综合征大鼠局部炎症反应。
Objective:To investigate the effect of internal heat needle treatment on the inflammatory reaction and Akt/NF-κB signaling pathway in damaged issues of rats with the third lumbar transverse process syndrome.Method:Ninety Sprague-Dawley rats were randomly divided into six groups:the model group,the normal group,drugs group,internal heat needle group,ordinary acupuncture group and warming acupuncture group,fifteen rats of each group.To establish the third lumbar syndrome in rats by embedding gelatin sponge,then give treatment after two weeks.After treatment,the blood samples were collected by jugular vein,and then the levels of TNF-α,IL-1β,IL-6 and IL-8 in serum were measured by ELISA.The histopathological changes of the third lumbar transverse process were observed by HE staining.The protein level of p-Akt(S473)and NF-κB(p65)was detected by Western blot.Results Compared with the normal group,the levels of TNF-α,IL-1β,IL-8 and IL-6 in serum of the model group were increased significantly,the inflammatory reaction in the injured tissues was serious,and the up-regulation of p-Akt(S473)and NF-kappa B(p65)protein was significant.Compared with the model group,the levels of TNF-α,IL-1β,IL-6 and IL-8 in each treatment group were decreased,the inflammation reaction of tissues was alleviated,the proteins expression of p-Akt(S473)and NF-kappa B(p65)were decreased,and the effect of internal heat needling group was the most obvious.Conclusion:Internal heat needle treatment can improve the local inflammatory response of the third lumbar transverse process syndrome,it may be related to its inhibition of Akt/NF-κB signaling pathway to reduce the expression of inflammatory cytokines.
