β-catenin参与乳腺癌的可能机制 被引量:6

摘要 乳腺癌是女性最常见的恶性肿瘤,其发病机制复杂。β联蛋白(β-catenin)是可表达于细胞膜和细胞质并参与生物体内多种重要生命活动的一种多功能蛋白,如参与组织发育分化、细胞间黏附迁移等,且在细胞内许多重要的信号通路转导均涉及β-catenin,如Wnt/β-catenin信号通路、第10号染色体缺失的磷酸酶及张力蛋白同源基因/蛋白激酶B/β-catenin信号通路;同时,β-catenin也受细胞内许多分子的调节,如N-myc下游调节基因、糖原合成酶激酶-3β等,如果这些信号通路转导或分子调节发生异常,则可能导致肿瘤的发生发展,包括乳腺癌。目前,关于β-catenin在乳腺癌、胃癌、结直肠癌等多种肿瘤形成演进中的作用机制已成为研究热点。未来,β-catenin是否可作为乳腺癌诊断、预后判断的分子标志物需进一步探索。 Breast cancer is the most common malignant tumor in women,and its pathogenesis is complex.β-catenin is a multifunctional protein which expresses on cell membrane and in cytoplasm and participtes in many important life activities,such as the development and differentiation of tissues,the adhesion and migration of cells,and many important signaling pathways,such as Wnt/β-catenin signaling pathway,phosphatase and tensin homologous genes deleted from chromosome 10/protein kinase B/β-catenin signaling pathway;meanwhile,β-catenin is regulated by many molecules in cells,such as N-myc downstream regulated gene 2,glycogen synthase kinase-3βand so on.If these signaling pathways′transduction and molecules′regulation are abnormal,it may lead to the occurrence and developmen of tumors,including breast cancer.The mechanisms ofβ-catenin in the formation and evolution of breast cancer,gastric cancer,colorectal cancer and other tumors has become a hot topic in oncology research.In the future,whetherβ-catenin can be used as a molecular marker for the diagnosis and prognosis of breast cancer needs to be further explored.
