摘要
山药炭疽病是危害山药最严重的病害之一,可导致30%~85%的产量损失。本研究采用柯赫氏法则对从病斑组织块分离的病原菌进行验证,通过形态学结合多基因分子系统学方法对病原菌进行鉴定。结果表明,分离获得的6株炭疽菌分别与喀斯特炭疽菌(Colletotrichum karstii)、胶孢炭疽菌(Colletotrichum gloeosporioides)、暹罗炭疽菌(Colletotrichum siamense)、隐秘炭疽菌(Colletotrichum aenigma)和兰科炭疽菌复合种(Colletotrichum orchidearum complex)的Colletotrichum plurivorum和Colletotrichum sojae聚在一起,支持率分别为89%、80%、87%、94%、88%和91%。这是关于喀斯特炭疽菌、暹罗炭疽菌、隐秘炭疽菌、C.plurivorum和C.sojae引起山药炭疽病的首次报道。致病性测试表明,6株菌株均能对山药离体叶片致病,但不同菌株的致病力具有一定差异。暹罗炭疽菌致病力最强,喀斯特炭疽菌、胶孢炭疽菌、C.plurivorum或C.sojae致病力中等,隐秘炭疽菌致病力最弱。山药炭疽病病原菌的种类构成和致病性分化存在差异,为深入研究该病发生规律、防治及抗病育种等工作提供理论依据。
Yam anthracnose is a serious pathogen,which causes a yield loss of 30%~85%.The pathogens were isolated by tissue associated anthracnose and confirmed through Koch’s postulates,the strains were identified through morphologic characteristics and multi-gene(ITS,GAPDH,CAL,ACT CHS-1,TUB2) combination analysis.The results showed that six strains to be the pathogenic agents that were identified as Colletotrichum karstii,C.gloeosporioides,C.siamense,C.aenigma,C.plurivorum and C.sojae.The bootstrap support of both clades were89%,80%,87%,94%,88% and 91%,respectively.This is the first report of C.karstii,C.siamense,C.aenigma,C.plurivorum and C.sojae for purple Dioscorea alata L.anthracnose.Pathogenicity tests showed that all of the isolates could infect detached leaves,with different pathogenicity for different strains.C.siamense presented the strongest virulence to yam leaves,followed by C.karstii,C.gloeosporioides,C.plurivorum and C.sojae,C.aenigma with weak pathogenicity.There are differences in species composition and pathogenic differentiation of anthracnose pathogens for yam,which will provide theoretical basis on further study disease regularity and control and breeding disease resistance yam cultivars.
